Correlation of hypoxia inducible factor 1-α polymorphism and nasopharyngeal carcinoma

Abstract: Objective To investigate the distribution of hypoxia inducible factor 1α(HIF-1α) C2028T polymorphism in patients with nasopharyngeal carcinoma(NPC) and its role in NPC carcinogenesis.Methods DNA genome from 90 patients with NPC were extracted,and the HIF-1α C2028T polymorphism was detected by PCR-RFLP method.The relations of HIF-1α C2028T polymorphism with the patients' clinicopathologic features were statically analyzed.Results There were only two genotypes,C/C and C/T.The proportions of genotype C/C and C/T were 87.8%(79/90)and 12.2%(11/90)respectively.The HIF-1α C2028T polymorphism was positively related to patients' N stage(rs=0.39,P=0.00),and was likely having a positive correlation with UICC stage(rs=0.21,P=0.05).There was no correlation between the polymorphism and pathological type(P=0.98) or T stage(P=0.88).Conclusions The genotype C/T of HIF-1α might improve the progress of NPC carcinogenesis,especially might improve the metastasis of NPC.Its exact mechanism deserves further study.
