Expression and prognostic significance of ERCC1 BAG-1 and BRCA1 mRNA in non-small cell lung cancer

Abstract: Objective To investigate the expression and prognostic usefulness of ERCC1,BAG-1 and BRCA1 in patients with non-small cell lung cancer(NSCLC) treated with platinum-based adjuvant chemotherapy. Methods RT-PCR was used to investigate the mRNA expression of ERCC1,BAG-1 and BRCA1 in 85 samples of NSCLC tissues and 34 tumor-adjacent tissues.Relationships of the expression of these three genes with overall survival(OS) were analyzed. Results ①OS was significantly longer in patients negative for ERCC1 or BAG-1 mRNA expression than in those positive for expression of either gene(P=0.001).Platinum-based chemotherapy was more effective for patients negative for ERCC1 or BAG-1 than for the corresponding positive patients(P=0.001 and P=0.002), respectively.②OS may be longer in patients negative for BRCA1 mRNA expression than in BRCA1-positive patients (P=0.052). ③Multivariate analysis suggests that ERCC1 and BAG-1 may be prognostic indicators for patients receiving platinum-based adjuvant chemotherapy(P=0.027 and P=0.022). Conclusion Expression of ERCC1 and BAG-1 mRNA may be useful in the prognosis of NSCLC treated with platinum.
