    Effects of Fufang Huazhuo Jiangqi on precancerous lesions of the esophagus in a mouse model

    Objective To observe the effects of the Fufang Huazhuo Jiangqi traditional Chinese medicine preparation on a mouse model of precancerous esophageal lesions. Methods Mice were treated with 4-nitro quinoline-oxide(4-NQO)to induce formation of precancerous esophageal lesions,and the animals were distributed into normal group,model group,positive control group,prevention group,treatment group Esophageal epithelial tissue morphology was examined by light microscopy at 14 weeks of treatment,and by electron microscopy at the end of the experiment. Results The model group showed greater cell volume, cell number,and nuclear size at 14 weeks,confirming the successful induction of precancerous esophageal lesions. Malignant transformation occurred in none of the animals in the normal group,80% of animals in the model group,21.4% in the positive control group,5.6% in the prevention group,and 12.5% in the treatment group. At the end of the experiment,animals in the model group showed intercellular broadening,desmosomes,nuclear pycnosis,and chromatin edges. Fewer pathological changes were observed in the positive control,prevention and treatment groups. Conclusion The Fufang Huazhuo Jiangqi prescription may significantly protect against malignant transformation of precancerous esophageal injury in a mouse model. It may therefore be useful for preventing esophageal carcinoma in humans.

    Association of Dnmt1 single-nucleotide polymorphisms and risk of gastric cancer

    Objective The aim of our study was to investigate whether single-nucleotide polymorphisms at loci rs16999593 and rs2228611 in the gene encoding DNA methyltransferase 1(Dnmt1) are associated with risk of gastric cancer. Methods A total of 310 patients with gastric cancer and 420 healthy controls,all Han Chinese, were genotyped at rs16999593 and rs2228611 using PCR-based DNA sequence analysis and direct DNA sequencing. Results TT,CT and CC genotypes were identified at the rs16999593 locus: respective frequencies among patients were 58.1%, 36.1% and 5.8%, compared to 66.9%, 27.9% and 5.2% among controls. The CT genotype was associated with significantly higher risk than the wild-type TT genotype(OR 1.494,95%CI 1.086 to 2.057, P<0.05). The CC genotype was associated with marginally,but not significantly,higher risk than TT(OR 1.277,95%CI 0.677 to 2.448,P>0.05). The frequencies of genotypes at rs2228611 did not differ significantly between patients and controls(P>0.05). Conclusion Polymorphism at Dnmt1 rs16999593 is associated with elevated risk of gastric cancer among Han Chinese in Qingdao.

    Expression and clinical significance of hTERT in gastric cancer

     Objective To investigate the expression and clinical significance of human telomerase reverse transcriptase (hTERT) in gastric cancer tissues. Methods Quantitative real-time PCR was used to measure levels of hTERT mRNA in gastric tissues,while MaxVisionTM  immunohistochemistry was used to detect expression of hTERT protein in gastric tissue sections. Possible correlations of hTERT mRNA and protein levels with clinicopathological characteristics and with patient survival were explored. Results The mRNA encoding hTERT was overexpressed in gastric caner tissues and adjacent tumor tissues(P<0.05),while the rate of hTERT protein-positive staining was significantly higher in gastric cancer tissues(83.3%) than in normal tissues (40.0%,P<0.05). Levels of hTERT mRNA correlated positively with protein levels(r=0.733,P<0.05). Up-regulation of hTERT was significantly associated with lymph node metastases,distant metastases and TNM stage(P<0.05). Kaplan-Meier survival analysis showed significantly higher survival rates for patients expressing low levels of hTERT than for those expressing high levels(P=0.015). Conclusions These results suggest that hTERT is overexpressed in gastric cancer tissues,and that such overexpression is associated with gastric cancer progression. Assaying hTERT levels may provide an indicator of disease progression and prognosis.

    Tumor stage and primary treatment selection among patients with hepatocellular carcinoma from 2003 to 2013

    Objective This study reviewed the frequencies of each tumor stage and each type of primary treatment modality among patients with primary hepatocellular carcinoma(HCC)treated at a tertiary tumor hospital over the last decade. Methods Baseline data of patients with primary HCC treated between January 2003 and October 2013 were retrospectively collected. Tumor stage was determined according to the Barcelona Clinic Liver Cancer(BCLC) staging system. Results A total of 6,241 patients with primary HCC were included in the analysis,of whom 28.9% were in stage BCLC-0/A;16.2%,BCLC-B;53.6%,BCLC-C;and 1.3%,BCLC-D. Treatment modalities applied to this patient group were as follows:hepatic resection,33.3%;interventional therapy,36.7%;radiotherapy,2.2%;ablation therapy,0.9%;systemic chemotherapy,8.8%;traditional herbal medicine therapy,4.2%;targeted drug therapy,0.1%;and no treatment, 13.8%.Hepatic resection was the most frequent therapy for patients with BCLC-0/A/B disease, and interventional treatment was the first-line therapy for patients with BCLC-C disease. Conclusion Prevalence of stage BCLC-B and-C disease was high among our HCC patients.Hepatic resection and interventional treatment are first-line therapies for both stages.

    Efficacy of bevacizumab combined with docetaxel to treat Her-2-negative metastatic breast cancer

    Objective To investigate the efficacy of bevacizumab in combination with docetaxel to treat Her-2-negative metastatic breast cancer. Methods  Patients received either bevacizumab in combination with docetaxel(n=42) or docetaxel(n=37). Curative outcomes were assessed after 4 cycles of therapy during follow-up lasting 3-24 months. Results The total efficacy rate was 59.52% in the group receiving bevacizumab and docetaxel and 37.84% in the group receiving docetaxel,which was a significant difference(P>0.05). The positive response rate was significantly higher in the group receiving bevacizumab and docetaxel (26.19 vs 24.32%,P<0.05),as was the 2-year survival rate(40.28 vs 16.22%,P<0.05). At the same time,incidence of anemia and thrombocytopenia was significantly higher in the group receiving bevacizumab and docetaxel. Conclusion Bevacizumab combined with docetaxel can show high efficacy against Her-2-negative metastatic breast cancer,as well as improve the survival rate and survival time. Future studies need to examine ways to reduce the adverse effects of the therapy on blood components.

    Prognosis analysis of gastrointestinal stromal tumor

    Objective  To investigate the histopathological features,immunohistochemical expression and prognostic factors of gas-trointestinal stromal tumor (GIST). Methods Clinical and pathological data were collected from 67 patients with gastrointestinal stromal tumors. All patients underwent routine pathological and immunohistochemical examination of CD117,while CD34,S-100,dog-100, desmin staining characteristics were also analyzed. Life-table method was used to estimate overall survival and survival plot curves. COX proportional hazards model was used to analyze the prognostic factors of GSIT. Results Of all the 67 cases,41 were located at stomach,17 were in bowel,and the other 9 were in the gastrointestinal tract. The most common seen histological type were fusiform common in 52 cases,followed by 12 cases of epithelial cell type,3 cases of mixed cell type. Immunohistochemistry results:61 were CD117-positive,52 were CD34-positive,65 weredog-1-positive,28 were S-100-positive and 23 were desmin-positive. The expression of  CD117,CD34,dog-1 were higher in the digestive tract than the other that are not(P<0.05). Multivariate analysis showed that GIST prognosis was associated with distant metastasis,Gleevec treatment and risk classification(P<0.05),but not associated with sex,age,tumor location, tumor size,and CD117 expression(P>0.05). Conclusions  The most common type of GIST was spindle cells and stomach is the most common occurrence site,followed by intestine. CD117,CD34,dog-1 were highly expressed in the GIST. Risk classification, metastasis and Gleevec treatment were independent prognostic factors of GIST.

    89SrCl2 combined with 99Tc-MDP in patients with bone metastases: aMeta-analysis

    Objective To evaluate the efficacy of 89SrCl2 combined with 99Tc-MDP for patients with bone metastases. Methods Randomized controlled trials published up to June 2015 that compared 89SrCl2  combined with 99Tc-MDP and 89SrCl2  alone for treating patients with bone metastases were retrieved from databases including the Cochrane Library,PubMed,EMBASE,WanFang,CBM,CNKI,and VIP. The quality of eligible studies was evaluated,and data were meta-analyzed using Revman 5.3. Results A total of 7 RCTs involving 691 patients were included in the meta-analysis. The combination of 89SrCl2  and 99Tc-MDP led to significantly higher efficacy in bone pain palliation(RR=0.16,95%CI:0.10-0.22),higher radionuclide bone scan score (RR= 1.31,95%CI:1.07-1.60) and higher KPS score(RR=1.68, 95%CI:1.33-2.12). The most frequent adverse reactions were leukopenia and thrombocytopenia. Conclusions The available evidence suggests that the combination of 89SrCl2  and 99Tc-MDP is superior to 89SrCl2 alone for palliating bone pain and improving radionuclide bone scan and KPS scores in patients with bone metastases. However,these findings should be confirmed and extended in future studies.

    Expression of NF-κB in B cell lymphoma carcinoma and its clinical significance

    Objective To investigate the expression of nuclear transcription factor κB(NF-κB)in B cell lymphoma,and analyze possible relationships between expression and clinicopathological characteristics. Methods Immunohistochemistry was used to detect NF-κB protein expression in tissues from 42 patients with B cell lymphoma and 20 patients with reactive lymph node hyperplasia. Relationships between NF-κB expression and clinicopathological characteristics were analyzed. Results The rate of NF-κB-positive tissue was significantly higher among patients with B cell lymphoma (78.57%,33/42) than among those with reactive lymph node hyperplasia(20%,4/20;P<0.001). Level of NF-κB protein expression correlated with Ann-Arbor clinical stage and histology grade (P<0.05),but not with sex,age or lactate dehydrogenase (LDH)level (P>0.05). Conclusion NF-κB plays an important role in the development of B cell lymphoma, and it may serve as a prognostic indicator.

    Analysis of Academic Influence of the Nine Core Oncology Periodicals in China for 2008-2012

    Objective To understand the present situation and trends in the influence of Chinese core journals in oncology. Methods Nine Chinese core journals in oncology were compared using 9 bibliometric indicators in the Chinese S&T Journal Citation Reports(2009~2013,Suppl),and a dynamic model was used to evaluate their academic influence. Results Over the past 5 years, impact factors,total cites and academic influence of the nine journals increased,respectively,by 7.67%,28.88% and 34.46%. The proportion of papers reporting research funding gradually increased over the same period. Conclusion The nine journals have increased in academic influence over the study period,although at different rates.
