肝癌耐药细胞株的建立及癌干细胞的特性

Establishment of a drug-resistant liver cancer cell line and its cancer stem cell characteris-tics

摘要: 目的 建立肝癌耐药细胞株,并观察耐药细胞株癌干细胞的相关生物学特性。方法 采用阿霉素(ADM)浓度梯度递增法诱导建立耐药BEL-7404人肝癌细胞株,并对亲代与耐药肝癌细胞株进行癌细胞基本特性、癌干细胞相关细胞学特性进行检测,用计数法检测癌细胞生长曲线、倍增时间与MTT法测耐药倍数;单个细胞成株培养、平板克隆形成实验、“球囊”细胞培养实验。结果 耐药BEL-7404人肝癌细胞与亲本细胞相似,呈上皮样生长方式,但其中梭状细胞略多,耐药倍数为292倍;生长曲线表明其增殖速度明显低于亲本BEL-7404人肝癌细胞,且倍增时间增加;耐药BEL-7404肝癌细胞克隆形成率、“球囊”细胞形成率均明显低于其亲本BEL-7404人肝癌细胞。结论 低浓度阿霉素持续增量法可诱导形成耐药肝癌细胞株,但耐药肝癌细胞株的癌干细胞特征并不明显。

Abstract: Objective To establish a drug-resistant liver cancer cell line and examine its cancer stem cell characteristics. Methods The drug-resistant cell line BEL-7404 was induced using a concentration gradient of adriamycin(ADM).Basic characteristics of the drug-resistant cell line were analyzed and compared to those of the parental cell line,such as cell growth,cell doubling time,cell cycle,resistance index(RI),single cell culture,colony formation and balloon cell culture. Results Similar to its parental cell line,the drug-resistant BEL-7404 line showed an epithelioid growth pattern,but with slightly more spindle cells.BEL-7404 cells had an RI of 292,and their cell proliferation rate was significantly slower than that of the parental cells,with increased cell doubling time and sig-nificantly lower rates of colony formation and balloon cell formation. Conclusions Sustained incremental exposure to low concentrations of ADM can induce a drug-resistant cell line,though the cancer stem cell characteristics of this line are not obvious.
