鲜壁虎活性组分对人肝癌细胞增殖和凋亡的影响

摘要： 目的 探讨鲜壁虎活性组分对人肝癌BEL-7404细胞增殖及凋亡的影响。方法 将鲜壁虎活性组分冻干粉配制为不同浓度的溶液，并作用于肝癌BEL-7404细胞。以MTT法检测不同浓度的鲜壁虎活性组分对肝癌BEL-7404细胞增殖的抑制作用，流式细胞术分析各组细胞的凋亡情况。结果 不同浓度的鲜壁虎活性组分均对肝癌BEL-7404细胞的增殖有抑制作用，且呈剂量-时间依赖效应，实验组的抑制率均较对照组显著提高（P<0.05）。不同浓度的鲜壁虎活性组分均能诱导肝癌BEL-7404细胞凋亡，高浓度组（2 560 μg/ml）的作用更为明显（P<0.05）。结论 鲜壁虎活性组分可显著抑制BEL-7404细胞的增殖，并诱导其凋亡。

Abstract: Objective To investigate the effects of fresh Gecko active component on proliferation and apoptosis in human hepatocel-lular carcinoma cell line BEL-7404. Methods Solutions were prepared containing different concentrations of freeze-dried fresh Gecko active component powder， and these were added to BEL-7404 cultures.MTT assay was used to measure the antiproliferative effects of the different concentrations，and flow cytometry was used to assess the extent of apoptosis at the different concentrations. Results Fresh Gecko active component inhibited BEL-7404 proliferation relative to the control group（P<0.05） in a time- and dose-dependent manner.The component also induced apoptosis in BEL-7404 cells，with the highest concentration（2 560 μg/ml） showing statistically significant effects（P<0.05）. Conclusion Fresh Gecko active component can efficiently inhibit cell proliferation and induce apoptosis in the human hepatocellular carcinoma cell line BEL-7404.
