姜黄素对急性早幼粒白血病细胞增殖分化及cofilin表达的影响

Effects of curcumin on acute promyelocytic leukemia cells:proliferation,differentiation and cofilin expression

摘要:

目的 探讨姜黄素对急性早幼粒白血病HL-60细胞(以下简称“HL-60细胞”)增殖、分化及丝切蛋白(cofilin)表达的影响。方法 以不同浓度(6.25 μmol/L、12.5 μmol/L、25 μmol/L)姜黄素作用于HL-60细胞,同时设立空白对照组。作用24 h后, 采用MTT法检测姜黄素对细胞增殖能力的影响,倒置相差显微镜观察细胞形态改变,流式细胞仪检测细胞周期变化,双向电泳与质谱分析检测cofilin蛋白表达。结果 不同浓度姜黄素作用24 h后,空白对照组细胞生长抑制率为0, 6.25 μmol/L组、12.5 μmol/L组和25 μmol/L组分别为(6.46±0.73)%、(9.79±1.36)% 和(79.48±7.99)%,与空白对照组比较差异均有统计学意义(P<0.05),且随药物浓度递增,抑制作用逐渐增强,呈浓度依赖性(P<0.05);倒置相差显微镜观察可见典型细胞生长阻滞的形态改变;流式细胞仪检测发现当姜黄素浓度为12.5 μmol/L时,细胞阻滞在G2 /M期;双向电泳与质谱分析发现姜黄素可下调HL-60细胞cofilin的表达。结论 姜黄素可在体外抑制急性早幼粒白血病HL-60细胞增殖,具有浓度依赖性,且在一定血药浓度时可使细胞周期发生阻滞,从而阻滞细胞分化,其作用机制可能与姜黄素下调细胞内cofilin的表达有关。

Abstract:

Objective To investigate the effects of curcumin on the proliferation and differentiation of HL-60 early immature leukemia cells as well as on their expression of silk protein(cofilin). Methods HL-60 cells were treated for 24 h with curcumin (6.25,12.5,and 25 μmol/L) or not (control). Then cultures were examined using the MTT assay to detect effects of curcumin on proliferation,inverted microscopy to detect changes in cell morphology,flow cytometry to detect alterations in the cell cycle,and two-dimensional electrophoresis and mass spectrometry to detect cofilin protein expression. Results The apoptotic rate of HL-60 cells increased with increasing curcumin concentration at 24 h,the rate in each concentration group were as follows:(6.46±0.73)%,6.25 μmol/L;(9.79±1.36)%,12.5 μmol/L;and(79.48±7.99)%,25 μmol/L,(P<0.05);they were higher with higher curcumin concentration(P<0.05).Inverted microscopy showed morphological changes typical for cell growth arrest. Flow cytometry showed that curcumin at 12.5 μmol/L arrested cells at the G2/M transition of the cell cycle. Two-dimensional electrophoresis and mass spectrometry analysis showed that curcumin down-regulated cofilin expression in HL-60 cells. Conclusion These in vitro studies suggest that curcumin can inhibit the proliferation of HL-60 promyelocytic leukemia cells in a concentration-dependent manner. Sufficiently high concentrations can arrest the cell cycle and block differentiation. At least some of these curcumin effects may occur via down-regulation of intracellular cofilin expression.
