CXCR4、PI3K和 AKT在三阴性乳腺癌细胞株MDA-MB-231中的表达

Expression of CXCR4,PI3K and AKT in MDA-MB?-231 triple-negative breast cancer cells

摘要:

目的 探讨CXCR4、PI3K及AKT在三阴性乳腺癌细胞株中的表达及其相关性。方法 采用过表达CXCR4质粒pcDNA3.1-CXCR4和沉默子siRNA-CXCR4分别转染三阴性乳腺癌细胞株MDA-MB-231,分为目的转染组和无关转染组,同时设立空白对照组。Western blot和qRT-PCR技术检测转染后CXCR4、PI3K、AKT 蛋白和mRNA的表达。结果 转染pcDNA3.1-CXCR4后,与空白对照组及无关转染组比较,目的转染组CXCR4、PI3K和AKT蛋白及mRNA表达水平升高(P<0.05);转染siRNA-CXCR4后,与空白对照组和无关转染组相比,目的转染组CXCR4、PI3K和AKT蛋白及mRNA表达水平下降(P<0.05)。结论 在三阴性乳腺癌细胞株MDA-MB-231中CXCR4可调节PI3K及AKT的表达。

Abstract:

Objective To explore the expression of CXC chemokine receptor-4 (CXCR4),phosphatidylinositol 3-kinase (PI3K) and AKT in the triple-negative breast cancer cell line MDA-MB-231. Methods MDA-MB-231 cells were transfected with pcDNA3.1 encoding CXCR4 or treated with siRNA against CXCR4. qRT-PCR and Western blotting were used to detect mRNA and protein expression of CXCR4,PI3K and AKT. Results Transfecting cells with pcDNA3.1-CXCR4 increased mRNA and protein levels of CXCR4,PI3K and AKT (P<0.05);siRNA-mediated silencing of CXCR4 reduced mRNA and protein levels of CXCR4,PI3K and AKT(P<0.05). Conclusion CXCR4 expression may regulate the expression of PI3K and AKT in triple-negative breast cancer.
