Effect of corydalis saxicola bunting aqueous extract on proliferation and migration of hepatocellular carcinoma HepG2 cells and its possible mechanism

Abstract:

Objective To investigate the effect of corydalis saxicola bunting aqueous extract on hepatocellular carcinoma HepG2 proliferation and migration and its possible mechanism. Methods The culture medium containing different concentrations(0.4 mg/mL 0.8 mg/mL 1.6 mg/mL) of corydalis saxicola bunting aqueous extract was applied to hepatocellular carcinoma HepG2 cells. The proliferation of HepG2 cells was assayed using the cell counting kit-8(CCK-8),and invasion was detected using the Transwell test. The expression of NF-κBp65 was detected by real-time PCR and Western blot analysis. Results The results of CCK-8 showed that the inhibition rate of HepG2 cells in different concentra-tions of corydalis saxicola bunting aqueous extract at 24 h,48 h,and 72 h was statistically significant (P<0.05),and the cell prolifer-ation in-hibition rate of 0.8 mg/mL and 1.6 mg/mL was higher than 0.4 mg/mL(P<0.05). The results of Transwell experiments showed that the cell migration of HepG2 cells treated with different concentrations of Rhizoma Coptidis aqueous extracts for 48 h was lower than that of the control group (P<0.05),and the expression of NF-Bp65 protein and mRNA was up-regulated (P<0.05). Conclusions Corydalis saxicola bunting aqueous extract inhibits proliferation,migration of HepG2,and the mechanism of it may do so by up-regulating NF-κBp65 expression.
