    Effect of NCLX-mediated mitochondrial calcium homeostasis on the growth of hepatocellular carcinoma cells

    Objective We performed this study to explore the effect of NCLX-mediated mitochondrial calcium on the growth of hep-atocellular carcinoma (HCC) cell line. Methods Eukaryotic expression vector pSliencer3.0-shNCLX and pcDNA3.1(+)-NCLX were transfected into HCC cells SNU-739 and SNU-368,respectively,and NCLX was stably knocked down or overexpressed. The expression level of NCLX protein was detected by Western blot assay. Rhod-2-AM staining was used to detect the level of mitochondrial calcium. The proliferation ability of cells was evaluated by MTS and EdU assay. The apoptosis of cells was assessed by apoptosis assay. Results Western blot assay showed that NCLX was stably knocked down or overexpressed in HCC cells SNU-739 and SNU-368. Compared with transfected empty vector,the mitochondrial calcium level was significantly increased after downregulation of NCLX expression (P<0.01),the cell proliferation rate was significantly increased (P<0.05),and the apoptosis rate was significantly decreased (P<0.01) in HCC cells SNU-739 and SNU-368. After upregulation of NCLX expression,the mitochondrial calcium level was significantly decreased (P<0.05),the cell proliferation rate was significantly decreased (P<0.05),and the apoptosis rate was significantly increased (P<0.01) in HCC cells SNU-739 and SNU-368. Conclusions In HCC cells,downregulation of NCLX-induced increase in mitochondrial calcium level may effectively promote cell proliferation and inhibit apoptosis. However,overexpression of NCLX-induced decrease in mitochondrial calcium levels inhibits cell proliferation and promotes apoptosis in HCC cells SNU-739 and SNU-368.These results suggest that NCLX may be a potential target for the treatment of hepatocellular carcinoma.

    Objective To investigate the effect of thrombospondin-2(THBS2) on proliferation,invasion and migration of human cervical squamous carcinoma SiHa Cells and the ability of tumor formation in nude mice. Methods Human cervical squamous  carcinoma SiHa cells transfected with the THBS2 overexpression  lentivirus served as the experimental group(THBS2-LV group),transfected with empty virus served as a negative control group (NC group) and untreated human cervical squamous  carcinoma SiHa cells served as a blank control group.The mRNA and THBS2 protein expression were detected by RT-qPCR and Western blot respectively. The proliferation and apoptotic rate of cells were assayed by CCK-8 method and flow cytometry respectively. Cell invasion and migration were analyzed by Transwell assay and Scratch test. The tumor formation ability of nude mice was observed by tumor formation experiment in nude mice. Results Compared with the NC group and the blank control group,the expression of THBS2 mRNA and protein and the apoptotic rate of cells increased in THBS2-LV group,while the cell proliferation,invasion and migration were reduced(P<0.05). The results of tumor formation in nude mice showed that the subcutaneous tumorigenic ability and average tumor volume of nude mice inoculated with THBS2-LV group were lower than those of nude mice inoculated with NC group and blank control group(P<0.01). Conclusions Overexpression of THBS2 can inhibit the proliferation,invasion,migration and subcutaneous tumor formation of cervical squamous carcinoma SiHa cells. Up-regulation of THBS2 can inhibit the development of cervical squamous cell carcinoma.

    Objective To investigate the effect of corydalis saxicola bunting aqueous extract on hepatocellular carcinoma HepG2 proliferation and migration and its possible mechanism. Methods The culture medium containing different concentrations(0.4 mg/mL 0.8 mg/mL 1.6 mg/mL) of corydalis saxicola bunting aqueous extract was applied to hepatocellular carcinoma HepG2 cells. The proliferation of HepG2 cells was assayed using the cell counting kit-8(CCK-8),and invasion was detected using the Transwell test. The expression of NF-κBp65 was detected by real-time PCR and Western blot analysis. Results The results of CCK-8 showed that the inhibition rate of HepG2 cells in different concentra-tions of corydalis saxicola bunting aqueous extract at 24 h,48 h,and 72 h was statistically significant (P<0.05),and the cell prolifer-ation in-hibition rate of 0.8 mg/mL and 1.6 mg/mL was higher than 0.4 mg/mL(P<0.05). The results of Transwell experiments showed that the cell migration of HepG2 cells treated with different concentrations of Rhizoma Coptidis aqueous extracts for 48 h was lower than that of the control group (P<0.05),and the expression of NF-Bp65 protein and mRNA was up-regulated (P<0.05). Conclusions Corydalis saxicola bunting aqueous extract inhibits proliferation,migration of HepG2,and the mechanism of it may do so by up-regulating NF-κBp65 expression.

    Effect of resveratrol on autophagy and apoptosis of non-small cell lung cancer cells

    Objective To investigate the effects of resveratrol (Res) on autophagy and apoptosis of non-small cell lung cancer A549 cells. Methods A549 cells were divided into control group and resveratrol group (Res group). Res group cells were treated with 50 μmol/L resveratrol,and the control group cells were cultured with DMEM containing 2% FBS. The morphological changes were observed by inverted microscopy. The formation of autophagosomes was observed by laser scanning confocoal. The expression of the autophagy-related proteins Beclin1,LC3Ⅱ/Ⅰ,p62 and apoptosis-related proteins Bcl-2,Bax,Active caspase-3 were detected by Western blot. Results After 24 h culture,the cell in the Res group were more round and sparsely arranged than those in the control group. A large number of autophagy bodies formed in the cytoplasm. Western blot showed that Beclin1 and and LC3Ⅱ/Ⅰ expression were increased in A549 cells in the Res group than in the control group(0.151±0.032 vs 0.093±0.013,P=0.011;3.644±0.122 vs 1.903±0.054,P<0.001),but p62 protein expression was reduced(0.032±0.002 vs 0.061±0.005,P=0.021) in the Res group. The expression of pro-apoptotic protein Bax and activated caspase-3 were increased in the Res group(0.633±0.061 vs 0.423±0.053,P=0.011;0.154±0.004 vs 0.111±0.011,P=0.002). However,the expression of anti-apoptotic protein Bcl-2 was reduced in the Res group(2.437±0.055 vs 3.503±0.138,P<0.001). Conclusion Resveratrol induces non-small cell lung cancer A549 cells apoptosis by promoting excessive autophagy.

    Correlation between HPV 16/18 infection and p53 expression in esophageal squamous cell carcinoma

    Objective To investigate the infection of HPV16/18 in esophageal squamous cell carcinoma (ESCC) and its relationship with p53 mutation. Methods PCR was used to detect HPV16/18 E6、E7 genes in 56 cases of ESCC tissues and 24 cases of normal esophageal tissues; immunohistochemical technique(SP) was used to detect the protein of p53. Results In total,the infection rates of HPV16/18 in ESCC tissue and normal tissue were 44.6% and 12.5%,respectively,and the positive expression rates of p53 were 41.1% and 4.2%,respectively. In ESCC tissue,patients displayed higher infection rate of HPV16/18 and positive expression rate of p53 as compared to those in the normal tissue(χ2=7.630,P=0.006; χ2=10.896,P<0.001).  Clinicopathological analysis demonstrated that the HPV16/18 infection in the ESCC tissue was associated with histopathologic grading,TNM stage and lymph node metastasis(P<0.05). Additionally, HPV16/18 infection rate increased in parallel with p53 positive expression rate (r=0.565,P<0.001). Kaplan-Meier survival analysis showed that the median OS of patients with positive HPV infection and positive p53 expression were less than those with negative HPV infection and negative for p53 expression(21 months vs 39 months,χ2=4.306,P=0.038;17 months vs 41 months,χ2=5.868,P=0.015). After adjustment for potential confounders,Multivariate Cox regression showed that HPV infection (HR=1.834,95% CI:1.010-3.330,P=0.046) and p53 positive expression (HR=2.189,95% CI:1.182-4.054,P=0.013) could increase the risk of death in patients with ESCC. Conclusion HPV infection and p53 positive expression were both higher in ESCC,which may jointly promote the occurrence and development of ESCC and affect its prognosis.

    Objective To explore the expression of Pygopus2 (PYGO2) and its clinical significance in breast cancer. Methods PYGO2 data were retrieved from the Oncomine database and the relationship between PYGO2 and prognosis of breast cancer patients was retrieved from an online survival analysis tool “Kaplan-Meier Plotter” database. Results A total of 45 studies (including 3,730 samples) were involved PYGO2 in breast cancer tissues and normal tissues. PYGO2 expression levels in breast cancer was higher than that in normal tissues(P<0.05). Kaplan-Meier Plotter of survival analysis indicated that high expression of PYGO2 was closely associated with better survival in breast cancer,including overall survival (HR=0.69,95%CI: 0.51-0.95,P=0.021),relapse-free survival (HR=0.60,95%CI: 0.51-0.70,P<0.001) and distant metastasis-free survival (HR=0.61,95%CI: 0.43-0.87,P=0.005). Conclusion PYGO2 is highly expressed in breast cancer tissues and associated with poor prognosis in breast cancer patients,which may be a potential biomarker for estimating the prognosis of breast cancer.

    Association of hyperthyroidism with the risk of breast cancer: a meta-analysis

    Objective  To systematically evaluate the correlation between hyperthyroidism and the risk of breast cancer,to provide the theoretical basis for etiological study and primary prevention of breast cancer. Methods  Data of case-control studies and cohort studies of hyperthyroidism and breast cancer were collected by searching 8 electronic databases from inception to April 2018. The Newcastle-Ottawa quality scale(NOS) was used to evaluate the quality of inclusion study and a Meta-analysis was conducted by RevMan 5.3 software. Results  Finally 17 studies were included in the meta-analysis,which including 10 case-control studies and 7 cohort studies,with a total of 66,116 cases. The concrete results were as follows:⑴case-control study:the results of unadjusted OR showed that hyperthyroidism couldn't increase the risk of breast cancer(OR=1.25,95%CI:0.92-1.70,P=0.150);the results of adjusted OR showed that hyperthyroidism could increase the risk of breast cancer(OR=1.11,95%CI:1.03-1.19),P=0.004).⑵cohort study:unadjusted OR and adjusted OR combined results showed that hyperthyroidism could increase the risk of breast cancer (unadjusted OR=1.17,95%CI:1.07-1.27,P<0.001;adjusted OR=1.11,95%CI:1.07-1.15,P<0.001). Conclusions Hyperthyroidism may increase the risk of breast cancer. However,the relationship between and breast cancer risk in different regions is not exactly the same.

    Comparison of efficacy of irinotecan  combined with raltitrexed or 5-FU  as second-line treatment for advanced colorectal cancer

    Objective  To compare the clinical efficacy of irinotecan plus raltitrexed and irinotecan plus 5-FU(FOLFIRI) in the second-line treatment of patients with advanced colorectal cancer. Methods A total of 60 patients with advanced colorectal cancer were enrolled after first-line chemotherapy failure. They were divided into observation group(irinotecan plus raltitrexed) and control group (FOLFIRI),30 patients in each group. The clinical efficacy and adverse reaction,over survival (OS) and time to progresion (TTP) were observed. Results There was no significant difference in the response rate between the observation group and the control group(26.67% vs 13.33%,χ2=1.67,P=0.20). The disease control rate in the observation group was higher than that in the control group,and the difference was statistically significant ( 63.33% vs 36.67%,χ2=4.27,P=0.04). The median OS and median TTP in the observation group were longer than those in the control group (9.9 months vs 7.5 months, χ2=4.461,P=0.012;5.5 months vs 3.7 months, χ2=6.735,P=0.022). The gastrointestinal advese reactions between the two groups were not significantly different (P>0.05). Conclusions Irinotecan combined with raltitrexed as the second line treatment showed satisfactory efficacy and safety to treat the  patients with advanced colorectal cancer. It can prolong survival and reduce adverse reactions compared with FOLFIRI,and it is worthy of clinical application.

    Effect of different ventilation modes on the expression of S100A8 protein in patients undergoing pulmonary lobectomy

    Objective To investigate the expression of Calgranulin A(S100A8) in serum during lobectomy under different ventilation modes. Methods Twenty patients with lung cancer who underwent pulmonary lobectomy were divided into one-lung ventilation group and two-lung ventilation group according to different ventilation modes,with 10 cases in each group. The operation time,anesthe-sia time,total infusion volume,oxygenation index,recovery time,extubation time,sputum suction times,pneumonia,atelectasis,hospi-tal stay and death were recorded. Western blot was used to detect the expression of S100A8 in the patients serum in the two groups before operation (T0),at the end of operation (T1),24 hours after operation (T2) and 48 hours after operation (T3). Results There were no significant differences in operation time,anesthesia time,total infusion volume,recovery time,extubation time,sputum suction times,pneumonia cases,atelectasis cases and death cases between the two groups(P>0.05). The oxygenation index in one-lung ventilation group was significantly lower than that in two-lung ventilation group [(296.20±77.64) mmHg vs  (319.20±30.23) mmHg,t=2.189,P=0.042],and the hospital stay increased significantly [(7.80±1.87) d vs (6.20±1.48) d,t=2.121,P=0.048]. There was no significant difference in the expression of S100A8 between one-lung ventilation group and two-lung ventilation group before operation(P>0.05). Repeated variance analysis showed that there was significant difference among the groups with different ventilation modes(F=310.76,P<0.001). The trend of change over time was different(F=24.85,P=0.025). There was interaction between the time and time point(F=18.85,P=0.001). Conclusions The intraoperative oxygenation index of lung cancer patients undergoing thoracotomy with one-lung ventilation is lower than that with two-lung ventilation,and the expression of S100A8 is higher,which may be related to the higher incidence of ventilator-associated lung injury.

    Objective To explore the effects of short-term structured psychological education intervention combined with Xiaoyaosan on moderate and severe depression,chemokines CCL2 and inflammatory factors IL-6 in patients with hepatocellular carcinoma during perioperative period. Methods 83 patients included with hepatocellular carcinoma accepted liver resection for the first time from October 2016 to February 2018. General information questionnaire,Hospital Anxiety and Depression Scale(HADS-D),and Patient Health Questionnaire-9 item (PHQ-9) were used to conduct the survey at baseline.Patients with moderate and severe depression were randomly divided into control group(n=42) and observation group(n=41) by SPSS software. The control group was treated with usual care combined with Xiaoyaosan,and the observation group was treated with short-term structural psychological education intervention on this basis. Repeated measures anova was used to compare the HADS-D,PHQ-9 scores and the level of CCL2,IL6 of the two groups at baseline,on day 5 post-surgery and 30 days after surgery. Results The time effect,intergroup effect and interaction effect of the HADS and PHQ-9 scores of the two groups were statistically significant (P<0.05). The HADS-D and PHQ-9 scores were significantly reduced at 30 d after surgery in the observation group compared with the control group (P<0.001),while the levels of CCL2,IL-6 were significantly lower in the observation group than control group at 30d after surgery(P<0.05). Conclusions Short-term structural psychological education intervention combined with Xiaoyaosan can effectively alleviate the moderate and severe depression in patients with hepatocellular carcinoma during perioperative period,and down-regulate the levels of CCL2 and IL-6 to promote the postoperative recovery of patients with hepatocellular carcinoma.
