    Biological effects of knocking down GLRX3 in nasopharyngeal carcinoma cells

    Objective To establish glutaredoxin 3(GLRX3)-knocked down nasopharyngeal carcinoma(NPC) cell lines and explore the biological functions of the GLRX3 gene. Methods Retroviral vectors expressing shRNA against GLRX3 were constructed and transfected into HONE1 cells, and stable cell lines showing knocked-down GLRX3 expression were selected. Reduced GLRX3 tran-scription in the cell lines was confirmed by real-time quantitative PCR.The cells were examined in colony formation and wound heal-ing assays to explore the influence of GLRX3 on these processes. Results Four retroviral shRNA vectors targeting GLRX3(shRNA-pBINNS2) were constructed and stably transfected into HONE1 cells.GLRX3 transcription was effectively suppressed in the stable cell lines.Knocking down GLRX3 inhibited clonogenicity and migration. Conclusions Knocking down GLRX3 inhibits the malignant behavior of NPC cells, suggesting that GLRX3 is a candidate oncogene in NPC.

    In vivo enrichment of liver cancer stem cells through adriamycin treatment:a preliminary study

    Objective To establish a nude mouse model of liver cancer and explore whether drug resistance of cancer stem-like cells in vivo can lead to enrichment of stem-like cells. Methods A subcutaneous tumor model of liver carcinoma was established by subcu-taneously injecting human liver BEL-7404 cell suspensions into both axilla of BALB/C-nu mice.Following adriamycin treatment,primary cultures were established, and both parental and adriamycin-treated cells were studied in assays of single-cell colony formation and sphere cell formation to detect drug resistance and analyze other cytological characteristics. Results The tumor formation rate in this BALB/C-nu subcutaneous tumor model was 100%.There were no significant differences in the rate of single-cell colony formation between BEL-7404 parental liver cells and first-generation cells after adriamycin treatment(P>0.05).However,second-generation cells showed a significantly higher rate of single-cell colony formation and a significantly lower colony formation rate than parental or first-generation cells(P<0.05). Non-adherent spheres were not observed in cultures of any of the three cell types. Conclusions Low-dose adriamycin intervention may be a way to enrich for liver cancer stem-like cells in vivo.

    Objective To explore the influences of the rs 12506899 and rs 16849384 polymorphisms in the α-fetoprotein(AFP) gene and the rs 7688274 polymorphism in the albumin(ALB) gene on AFP levels in human hepatoma cell lines. Methods AFP levels in the medium were measured for cultures of AFP-negative normal human cell line HL-7702 and AFP-positive human hepatoma cell lines BEL-7402,BEL-7404,Hep-G2,MHCC-97H and SMMC-7721.DNA was extracted and genotyped by real-time PCR(Taq-man assay). AFP levels were compared for hepatoma cell lines with different genotypes. Results A statistically significant association was observed between rs 16849384 and AFP levels.AFP levels were higher in Hep-G2 cells carrying a T/T genotype at rs 16849384 than in SMMC-7721,HL-7702 and BEL-7402 cells carrying a C/T genotype(P<0.001,<0.001 and<0.015,respectively). Conclusions Genotypes of rs 16849384 may be associated with AFP levels in hepatoma cell lines,and the T allele of rs 16849384 is associated with higher AFP level.

    Correlation between microRNA-146a polymorphism and primary liver carcinoma in the Guangxi Zhuang population

    Objective To investigate the correlation between the microRNA-146a rs 2910164 polymorphism and primary liver carcinoma(PLC) in a Guangxi Zhuang population. Methods Genotypes and alleles of rs 2910164 were determined in 110 PLC patients and 110 healthy controls in a Guangxi area with high PLC incidence using matrix-assisted laser desorption ionization time-of-flight mass spectrometry(MALDI-TOF MS). Differences in the distribution of polymorphisms between patients and controls were analyzed by the chi-squared test.Correlation between the rs 2910164 polymorphism and PLC risk was evaluated by non-conditional logistic regression. Results The CG genotype was significantly more frequent in PLC patients than in controls(P=0.026),and it was associated with increased risk of PLC(OR=3.473,95%CI 1.116-10.802,P=0.032).However,the frequency of the GG genotype was similar between the two groups(P>0.05).Similarly,the frequencies of the C and G alleles were similar in patients(62.7% and 37.3%,respectively) and controls(67.7% and 32.3%,P>0.05). Conclusions Carriers of the microRNA-146a rs 2910164 CG genotype are at elevated risk of PLC.This genotype may be a PLC risk factor in the Guangxi Zhuang population.

    Effects of trimetazidine on myocardial energy metabolism after adriamycin chemotherapy in a nude mouse breast cancer model

     Objective To investigate the myocardial protective effects of trimetazidine(TMZ) and its mechanism of action by re-searching the drug′s influence on myocardial energy metabolism after adriamycin(ADM) chemotherapy in a nude mouse breast cancer model. Methods Nude mice(n=27) with breast cancer were randomly divided into three groups:group A(control group) received saline(10 ml/kg) injections into the abdominal cavity,and groups B and C received ADM injections(2.5 ml/kg) into the abdominal cavity twice a week for 4 weeks. From the third week,group C was given TMZ by gavage 20 mg/(kg·d),once a day for 14 days; groups A and B received the same amounts of saline at the same times. Mice were anesthetized on the second day after the last administration.At three time points(before gavage,T1;before anesthesia,T2;30 min afer anesthesia,T3),blood(0.2 ml) and myocardial tissue specimens were taken from mice in all three groups.Serum levels of cTnI and activity of myocardial mitochondrial long-chain 3-ketoacyl coenzyme A thiolase(3-KAT) were measured by enzyme-linked immunosorbent assay(ELISA).Levels of adenosine triphosphate(ATP) and adenosine diphosphate(ADP) in myocardial tissue were determined by high performance liquid chromatography(HPLC). Results  ① At T1,serum cTnI level was higher in groups B and C than in group A(P<0.05),but there was no significant difference between groups B and C(P>0.05).At T2 and T3,the cTnI level was significantly lower in group C than in group B(P<0.05).Serum cTnI level in group A did not vary significantly across all three time points(P>0.05).In group B,in contrast,serum cTnI level was higher at T3 than at T1 and T2(P<0.05).In group C,the cTnI level was significantly higher at T1 than at T2 or T3,and T2 was significantly lower than at T1 and T3(P<0.05).② ATP and ADP levels were significantly different among the three groups(P<0.05);these levels were higher in group C than in group B(P<0.05).③ 3-KAT levels were significantly different among the three groups(P<0.05);the level was lower in group C than in group B(P<0.05). Conclusion  Trimetazidine protects against myocardial injury induced by ADM chemotherapy in a nude mouse breast cancer model.It exerts these protective effects by improving myocardial energy metabolism,inhibiting myocardial mitochondrial 3-KAT activity and improving ATP and ADP levels in myocardial tissue.

    Expression of AKR1B10 in hepatocellular carcinoma and its relationship with AFP level

    Objective To explore the expression of aldo-keto reductase family 1 member B10 (AKR1B10) in hepatocellular carcinoma (HCC)and its relationship with AFP level. Methods The expression of AKR1B10 protein was examined immunohistochemically in 65 cases of human HCC and adjacent liver tissues,as well as in 14 cases of normal human liver tissue.Levels of AKR1B10 pro-tein expression in HCC and preoperative serum AFP level were analyzed for a possible relationship. Results The expression rate and level of AKR1B10 protein were up-regulated in human HCC samples compared to the corresponding adjacent liver tissues,and in adjacent liver tissues compared to normal liver tissue(all P<0.05).Up-regulated AKR1B10 expression was found in 50 of 65 HCC cases (76.9%), higher than the proportion of 55.4% of patients with detectable preoperative serum levels of AFP.Moreover,AKR1B10 protein was detected in 21 of 29 HCC cases (72.4%) that were negative for preoperative serum AFP.Statistical analysis showed that AKR1B10 pro-tein expression in HCC tissue was not correlated with preoperative serum AFP level.      Cloning of SATB1 cDNA and subcloning into a eukaryotic expression vector

    Objective To clone the cDNA of the gene encoding special AT-rich sequence binding protein (SATB1) and to subclone it into eukaryotic expression vector pEGFP in order to investigate the function and mechanism of action of SATB1. Methods SATB1 cDNA was cloned from pCMV6-XL6-SATB1 by polymerase chain reaction (PCR) and then the target gene was subcloned into pGEM-T and subsequently into the eukaryotic expression vector pEGFP-N1 to give pEGFP-SATB1. Results The complete cDNA sequence of SATB1(2312 bp) was consistent with the reported sequence(Genbank BC001744.1).Sequencing and restriction digest analysis re-vealed the target gene to be inserted in the expression vector in the correct orientation. Conclusion The SATB1 cDNA was success-fully cloned and used to construct recombinant eukaryotic expression vector pEGFP-SATB1.

    Quantitation of alpha fetoprotein (AFP) in human serum using liquid chip technology

    Objective To develop liquid chip-based method for assaying alpha fetoprotein(AFP)in human serum and to evaluate method performance. Methods Different amounts of anti-AFP monoclonal antibody were coupled to fluorescent beads using liquid chip technology in a sandwich immunoassay format. Fluorescent microspheres prepared with the optimal amount of anti-AFP antibody were reacted with different concentrations of biotin-goat anti-mouse IgG,and several performance indicators were measured,including linear range,detection limit,and accuracy.The method was used to measure serum AFP concentration in 134 patients with hepatocellular carcinoma(HCC) and 47 healthy controls,and the results were compared with those obtained by electrochemoluminescence immunoassay(ECLIA). Results Optimal coupling was obtained with 8 μg of anti-AFP monoclonal antibody and 4 μg/ml of biotin-goat anti-mouse IgG.The linear response range for the method to detect serum AFP was 1.31~168 ng/ml and the detection limit was 0.424 ng/ml. Intra- and inter-assay coefficients of variation ranged,respectively,from 4.33% to 7.66% and from 9.82% to 13.85%.The method detected significantly diffe-rent serum AFP levels between patients with HCC and healthy controls(P<0.05).Method sensitivity was 85.82% and specificity was 95.74%.Measurements obtained by the liquid chip method showed a correlation coefficient(r) of 0.845(P<0.05) with measurements obtained by ECLIA. Conclusion The liquid chip and ECLIA methods showed good correlation in detecting AFP.Since the liquid chip method offers a wider detection range and greater sensitivity than ECLIA,in addition to showing good repeatability,it may allow faster detection with fewer samples.This approach may also prove useful for detecting AFP together with other multiplexing markers in HCC.

    Correlation between MDR1 polymorphism and primary liver cancer in Guangxi

    Objective To evaluate the correlation between the C1236T rs 1128503 polymorphism in the multidrug resistance 1 (MDR1) gene and primary liver cancer (PLC) in a Guangxi Zhuang population. Methods A case-control study of 109 PLC patients and 109 healthy controls was conducted in a Guangxi area with a high PLC incidence.Genotypes and alleles of MDR1 C1236T rs 1128503 were detected by matrix-assisted laser desorption ionization time-of-flight mass spectrometry(MALDI-TOF-MS). Results The PLC and control group did not differ significantly in frequencies of MDR1 rs 1128503 CC,CT,or TT genotypes,or in frequencies of C and T alleles(P>0.05).The frequencies of C and T alleles were 42.20% and 57.80%. Conclusions Risk of PLC is not increased in carriers of the MDR1 C1236T rs 1128503 CT or TT genotypes.MDR1 polymorphism may not be a genetic susceptibility factor for PLC.

    Inhibitory effects of metformin on breast cancer cell line MCF-7

    Objective To explore the mechanism of metformin in inhibiting breast cancer cell line MCF-7. Methods MCF-7 cells were cultured for 48 h in the presence or absence of 16.56 g/L metformin,total protein was extracted from cells and expression of nu-clear factor-kappa B(NF-κB) p65 and Lin28B were detected by Western blot. Furthermore, transcription levels of HMGA2 and H-Ras, which are regulated by microRNA let-7,were determined by RT-PCR. Results NF-κB p65 and Lin28B expression declined signifi-cantly after 48-h drug treatment(t=23.44,t=14.24;P<0.05).Transcription levels of HMGA2 and H-Ras,which are related to self-renewal and multipotent differentiation of tumor cells,decreased 85%(P<0.05) and 96%(P<0.05),respectively,based on 2-△△Ct measurements. Conclusions Metformin at 16.56 g/L can effectively reduce NF-κB and Lin28B expression and inhibit HMGA2 and H-Ras transcrip-tion in MCF-7 cells.These findings suggest that metformin inhibits breast cancer at the cellular level.

    Expression and clinical significance of IGFBP4 and PAPPA in non-small cell lung cancer

    Objective To investigate the expression and clinical significance of IGFBP4 and PAPPA in patients with non-small cell lung cancer(NSCLC). Methods Total RNA was extracted from 55 pairs of NSCLC tissues and adjacent noncancerous tissues.IGFBP4 and PAPPA expression was detected using real-time quantitative RT-PCR. Results IGFBP4 expression was significantly lower in NSCLC tissue than in adjacent lung tissue,but significantly higher than in normal lung tissue(P<0.05).IGFBP4 expression was significantly higher in squamous carcinoma tissues than in adenocarcinoma.Furthermore,survival was longer in patients with high IGFBP4 expression than in those with low expression(P<0.05).However,IGFBP4 expression did not correlate with age,gender,histological dif-ferentiation grade or lymph node metastasis.PAPPA expression was significantly higher in NSCLC tissue than in adjacent or normal lung tissue(P<0.05).PAPPA expression was higher in cases of adenocarcinoma or lymph node metastasis than in cases of squamous carcinoma or cases without lymph node metastasis(P<0.05).Survival was shorter in patients expressing high levels of PAPPA than in those expressing low levels(P<0.05).However,PAPPA expression was not related with age,sex or clinical stage.There was a signifi-cant positive correlation between IGFBP4 and PAPPA expression in NSCLC tissues(r=0.512,P=0.000). Conclusions IGFBP4 and PAPPA may correlate with tumor growth in NSCLC,and may be useful as molecular prognostic indicators of NSCLC.

    Comparison of different beam arrangements in intensity-modulated radiation therapy to treat nasopharyngeal carcinoma

    Objective To compare different beam arrangements in 7-field intensity-modulated radiation therapy(IMRT) to treat nasopharyngeal carcinoma by analyzing doses delivered to target tissue and adjacent normal tissue. Methods A total of 12 patients with nasopharyngeal carcinoma who had been prescribed the same total IMRT dose were treated with one of the following three IMRT plans:average plan,with a beam angle interval of 51°;anterior plan,with a beam angle interval of 42°;and a non-coplanar plan,involving 5 coplanar fields with a beam angle interval of 72°,as well as 2 non-coplanar fields with a couch angle of 90° and gantry angles of 30° and 330°. All plans were optimized using the same constraints to ensure comparability of the results.The following out-comes were measured:dose distribution to the planned target volume(PTV) and to organs at risk,dose volume histogram,PTV confor-mity index,mean dose(Dmean),minimum dose,and maximum dose(Dmax). Results The three IMRT plans provided similar dose distributions to the PTV.The anterior plan provided a slightly higher Dmax-1%(D1) to the spinal cord and brain stem than did the other plans,but this difference was very small and within the range of tolerated doses.The average plan provided a temporal lobe dose D33 that was 9.28% smaller than that of the anterior plan and 12.38% smaller than that of the non-coplanar plan.The non-coplanar plan provided a lower D50 to the parotid gland and temporomandibular joint than did the other plans,and the non-coplanar plan provided a temporomandibular joint dose D50 that was 30.47% smaller than that of the average plan and 33.29% smaller than that of the anterior plan.The non-coplanar plan provided an oral mucosa Dmean that was 10.49% higher than that of the average plan and 10.85% higher than that of the anterior plan. Conclusion Satisfactory dose coverage in the nasopharyngeal carcinoma target volume was achieved using 7-field IMRT.By designing individualized IMRT plans based on the location and volume of the target area, and by optimizing the type of beam arrangement,the dose distribution can be improved and the risk of affecting normal tissue signifi-cantly reduced.

    Objective To evaluate the diagnostic value of diffusion weighted imaging (DWI) for assessing radiation damage to parotid glands of miniature pigs. Methods Miniature pigs were exposed to single-dose radiation on either the left or right parotid gland(n=8) or they were not exposed to radiation(n=4),then all pigs were examined by conventional magnetic resonance imaging and DWI.Pathological changes and ADC values for parotid glands were recorded before and after irradiation. Results Before irradiation,the three groups did not differ significantly in salivary flow rate or ADC value(P>0.05).The flow rate increased slightly in the control group over the course of the study,and both flow rate and ADC value were significantly lower in the irradiated groups than in the control group (P<0.05).In addition, the two groups irradiated on the left or right parotid differed significantly in flow rate and ADC value(P<0.05). Conclusions The ADC value may indicate microstructural changes in parotid gland tissue after irradiation,and DWI may prove a useful method for monitoring parotid gland radiosensitivity.

    Expression and clinical significance of EZH2,FHIT and VEGF in bladder urothelial car-cinoma

    Objective To investigate the expression and clinical significance of EZH2,FHIT and VEGF in primary bladder urothe-lial carcinoma(BUC). Methods Tissue samples of primary BUC(n=76) and normal bladder(n=10) were assayed by immunohisto-chemistry. Results EZH2 and VEGF expression rates were much higher in BUC tissue than in normal bladder tissue(P<0.05),while the FHIT expression rate was significantly lower in BUC(P<0.05).Expression of EZH2,FHIT and VEGF was related to histological classification,pathological grade and clinical TNM stage of BUC(P<0.05). Conclusions Expression of EZH2,FHIT and VEGF was closely related to the histological classification,pathological grade and clinical TNM stage in BUC tissue. EZH2,FHIT and VEGF may play important roles in the pathogenesis of BUC.

    Diagnostic value of ultrasound elastography for detecting malignant cervical lymph nodes

    Objective To assess the diagnostic value of ultrasound elastography(UE) for detecting malignant cervical lymph nodes. Methods A total of 96 patients with cervical lymph nodes were analyzed by UE and by conventional ultrasonography(US) involving two-dimensional ultrasonography and color Doppler flow imaging. An elastic method was used to evaluate the diagnostic value of UE. All malignant lymph nodes were confirmed by pathology,and the diagnostic performance of the two methods was compared. Results The sensitivity,specificity,accuracy,and positive and negative predictive values of US in diagnosing malignant cervical lymph nodes were,respectively,76.6%,81.3%,78.1%,89.1% and 63.4%.The corresponding values for UE were 90.6%,81.3%,87.5%,90.6% and 81.3%.UE showed higher accuracy than US for diagnosing malignant cervical lymph nodes.The diagnostic agreement rate of UE with surgical pathology was 0.540(P<0.001),compared to 0.719(P<0.001) for US.UE showed higher diagnostic consistency than US. Conclusion UE may be useful for diagnosing malignant cervical lymph nodes.

Methods A total of 203 cases of nasopharyngeal carcinoma with cervical lymph node metastasis were randomly divided into two groups:the control group(98 cases),which was treated with  combined 5-fluorouracil and cisplatin chemotherapy,as well as routine radiotherapy; and the hyperthermia group(105 cases),which was treated with the same chemo- and radiotherapy as the control group to treat nasopharyngeal carcinoma,while cervical lymph node metastasis was treated with hyperthermia accompanied by radiotherapy. Results Complete response rates of cervical lymph nodes were 62.2% in the control group and 81.0%  in the hyperthermia group(P<0.05).The 5 year disease-free survival rates was 20.4% in the control group and 51.4% in the hyperthermia group(P<0.05).The 3- and 5-year overall survival rate were 52.0% and 40.8% in the control group and 76.2% and 58.1% in the hyperthermia group(P>0.05). Conclusions The combination of chemotherapy-radiotherapy-hyperthermia is more effective than chemotherapy-radiotherapy for patients with nasopharyngeal carcinoma and cervical lymph node metastasis.

    Effects of a combined Rituximab and CHOP regimen in patients with diffuse large B-cell lymphoma

    Objective To evaluate the efficacy and toxicity of rituximab plus CHOP in patients with diffuse large B-cell lymphoma. Methods A total of 48 patients with histopathologically confirmed CD20-positive diffuse large B-cell lymphoma were randomly as-signed into two groups:24 received CHOP plus rituximab(R-CHOP group),and 24 received CHOP alone(CHOP group).All patients received six chemotherapy cycles,and the therapeutic effect was assessed. Results The overall response rate was 91.7%(22/24) in the R-CHOP group and 66.7%(16/24) in the CHOP group(P<0.05).The 1-and 3-year survival rates were 95.8% and 83.3% in the R-CHOP group,and 74.7% and 50.0% in the CHOP group.The 1-and 3-year progression-free rates were significantly higher in the R-CHOP group(82.6% and 62.8%) than in the CHOP group(57.3% and 37.2%, P<0.05).The major side effects in the two groups were myelosuppression,gastrointestinal toxicity and infusion-related adverse reactions.The two groups did not differ significantly in adverse reactions, except chills and fever. Conclusion Combination chemotherapy including Rituximab and CHOP(R-CHOP) is effective and well-tolerated by patients, suggesting it is a good treatment for diffuse large B-cell lymphoma.

    Prognostic analysis of patients with primary retroperitoneal tumors undergoing surgical resection

    Objective To explore the prognostic factors of patients with primary retroperitoneal tumor(PRT) treated by surgical re-section. Methods We retrospectively analyzed 80 patients with PRT treated in the First Affiliated Hospital of Guangxi Medical Uni-versity between June 2001 and June 2012.Survival and prognosis were analyzed using the Kaplan-Meier method,the Cox proportional hazard model and the log-rank test. Results All patients were followed up for a median of 40.4 months, and the 1-, 3-, and 5-year cumulative survival rates were 92.5%,73.1% and 67.5%.Median survival time was 67 months. Univariate analysis identified the fol-lowing significant predictors of survival(P<0.05): surgical method,tumor size,tumor characteristics,mitotic counts of tumor cells,and invasion of adjacent organs.Multivariate analysis identified the following independent prognostic factors(P):surgical method,tumor <0.05characteristics,mitotic counts of the tumor cells and invasion of adjacent organs. Conclusion Surgical resection is the most ef-fective treatment for PRT.Tumor characteristics, surgical method,invasion of adjacent organs and mitotic counts of tumor cells are the most important prognostic factors for PRT.

    Analysis of health behavior self-efficacy in gastrointestinal cancer patients and factors that influence it

    Objective To investigate health behavior self-efficacy among gastrointestinal cancer patients after surgery and chemotherapy,and to analyze factors that influence it. Methods A total of 586 gastrointestinal cancer patients filled out a question-naire about self-efficacy,demographic characteristics and clinical disease information. Results The overall score of health behavior self-efficacy was 71.50 ± 16.08 for patients.Only 132(22%) patients scored over 80%.Patient nutritional status,education and medical care positively correlated with general self-efficacy(P<0.05),while age,clinical stage,chronic disease,stoma and metastasis negatively correlated with it(P<0.05).Stoma,chronic disease,nutritional status and age were the main predictors of self-efficacy. Conclusions Health behavior self-efficacy is still at a low level in most gastrointestinal cancer patients,which should be taken into account when designing and implementing clinical care.Efforts are needed to help patients adopt and maintain a health-promoting lifestyle in order to improve their quality of life.
