    LncRNA DLX6⁃AS1 affects the malignant biological behavior of esophageal cancer cells through the miR⁃181b/IL⁃6 axis

    Objective To investigate the effect of long non-coding RNA (lncRNA) homeobox transcription factor 6 antisense RNA1 (DLX6-AS1) regulating miR-181b/interleukin-6 (IL-6) axis on the proliferation, migration and invasion of esophageal cancer Eca-109 cells. Methods The human esophageal epithelial cell line HEEC and human esophageal squamous cell carcinoma cell line Eca-109 were selected, and the expression levels of lncRNA DLX6-AS1, miR-181b and IL-6 were detected by qRT-PCR. The si-DLX6-AS1, miR-181b mimics, pcDNA-IL-6, pcDNA-IL-6+miR-181b mimics and their corresponding negative controls were transfected into Eca-109 cells, respectively, and the transfection effect was detected by qRT-PCR. The CCK-8 assay and EdU staining assay were used to detect cell proliferation ability. The Transwell assay was used to detect cell migration and invasion ability, and dual luciferase reporter assay was used to verify the targeting relationship between lncRNA DLX6-AS1 and miR-181b, miR-181b and IL-6. Results Compared with those of HEEC cells, the expression levels of lncRNA DLX6-AS1 and IL-6 mRNA were increased in Eca-109 cells (P=0.005, 0.006), but the expression level of miR-181b was decreased (P=0.004). Compared with the corresponding negative control group, the cell proliferation activity and EdU positive cell rate in the si-DLX6-AS1 group and miR-181b mimics group were decreased (all P<0.05), and the number of cell migration and invasion were decreased (all P<0.05 ), while the cell proliferation activity and EdU positive cell rate in the pcDNA-IL-6 group were increased (both P<0.05), and the number of cell migration and invasion were increased (both P<0.05). IncRNA DLX6-AS1 had a targeted binding relationship with miR-181b. After co-transfection of miR-181b mimics and wild-type DLX6-AS1 vector, the intracellular luciferase activity was significantly decreased in cells (P=0.006). The expression level of miR-181b in Eca-109 cells was increased after down-regulation of DLX6-AS1 expression (P=0.010). miR-181b had a targeted binding relationship with IL-6. After co-transfection of miR-181b mimics and wild-type IL-6 vector, the intracellular luciferase activity was significantly decreased (P<0.05). The expression level of IL-6 in Eca-109 cells decreased after overexpression of miR-181b mimics (P<0.05). Compared with those of the pcDNA-IL-6 group, the expression levels of IL-6 mRNA and protein in Eca-109 cells, the cell proliferation activity, the EdU positive cell rate, the number of cell migration and invasion were decreased (all P<0.05), after co-transfection of pcDNA-IL-6 and miR-181b mimics. Conclusions LncRNA DLX6-AS1 promotes the proliferation, migration and invasion of esophageal cancer Eca-109 cells through miR-181b targeting and negatively regulating IL-6 expression.

    Effect of Apatinib on radiosensitivity of gastric cancer HGC⁃27 cells and its mechanism

     Objective To investigate the effect of Apatinib on the radiosensitivity of gastric cancer HGC-27 cells and its possible mechanism. Methods HGC-27 cells were treated with different concentrations of Apatinib (5 μmol/L, 10 μmol/L, 15 μmol/L, 20 μmol/L) and different irradiation doses (2 Gy, 4 Gy, 6 Gy, 10 Gy, 20 Gy). HGC-27 cells were then irradiated with 2 Gy and 6 Gy alone or combined with 10 μmol/L Apatinib. The expression of vascular endothelial growth factor (VEGF) was detected by ELISA, cell proli-feration capacity was detected by CCK-8, cell apoptosis and cell cycle were detected by flow cytometry, and expression of γ-H2AX was detected by immunofluorescence staining. Results Apatinib inhibited the proliferation of gastric cancer HGC-27 cells in a concentration- and time-dependent manner (all P<0.05). Different doses of irradiation promoted the release of VEGF in a time-and dose-dependent manner (all P<0.01). After the combination of 10 μmol/L Apatinib with 2 Gy and 6 Gy irradiation, respectively, the inhibition of cell proliferation, apoptosis rate and proportion of cells in G2/M phase of HGC-27 cells were increased compared with those in the single irradia-tion group (all P<0.01). The effect intensity of 6 Gy combined group was greater than that of 2 Gy combined group (all P<0.01). The nucleus focal quenching of γ-H2AX in the 6 Gy combined group was delayed compared with that in the 6 Gy single irradiation group. Conclusions Apatinib enhances the radiosensitivity of gastric cancer HGC-27 cells by inhibiting cell proliferation, promoting cell apoptosis and inducing cell cycle redistribution. The underlying mechanism may be related to the delay of γ-H2AX expression and the interference of DNA double-strand fracture repair.

    Analysis of the characteristics of diffuse gastric cancer and expression of hub gene LUM based on transcriptome sequencing

     Objective To analyze the transcriptome signatures of diffuse gastric cancer (DGC) and the expression of hub gene LUM (Lumican). Methods A total of 29 gastric cancer (GC) tissue and their corresponding adjust normal tissue samples were collected from patients who underwent GC resection between January 2021 and December 2021 at Guangxi Medical University Cancer Hospital, of which 5 were used to obtain mRNA expression profiling of DGC by RNA sequencing (RNA-Seq). Differentially expressed genes (DEGs) were analyzed by GO and KEGG. The protein-protein interaction (PPI) network of DEGs was constructed by cytoscape, and then the dominant gene set was evaluated. Based on the transcriptome and clinical data of GC in TCGA database, the gene expression and the prognosis were analyzed. RT-qPCR was used to examine the expression of hub gene LUM in other 24 GC tissues. Results Compared with normal adjacent tissues, a total of 175 DEGs in DGC, of which 91 genes were up-regulated and 84 genes were down-regulated. GO and KEGG enrichment analysis revealed that there were multiple ways involved in extracellular matrix related cellular processes and malignant progression in DGC. The EMT-associated gene set was dominant in DGC. The expression levels of COL6A3, LUM, COL1A2, SPARC, COL1A1, CDH11, COL3A1, NID2, BGN and COL4A1 in the EMT-associated gene set were higher than those in the adjacent normal and intestinal GC (all P<0.05). DGC patients with high expression of the hub gene LUM had a shorter overall survival (P=0.043). The RT-qPCR results showed that LUM was highly expressed in GC, and the expression level in DGC was higher than that in the intestinal and mixed GC(all P<0.05). Conclusions The transcriptome characteristics of DGC shows that EMT-associated gene set is in a dominant position. The expression of hub gene LUM is increased in DGC and the overall survival of patients with high LUM expression is shortened. LUM can be used as a potential prognostic marker and therapeutic target for DGC.

    Relationship between prognosis and the expression of H3K27M and its methylation in diffuse spinal glioma

    Objective To investigate the expression of histone H3 lysine 27-to-methionine mutations (H3K27M) and its methylation in diffuse spinal glioma and its relationship with prognosis. Methods A total of 75 patients with diffuse spinal glioma admitted to Zhongnan Hospital of Wuhan University from January 2016 to January 2019 were selected as the research subjects. The expression of H3K27M and H3K27M trimethylation (H3K27me3) in glioma tissues was detected by immunohistochemistry and chromatin immunoprecipitation combined with microarray assay. The multivariable Cox regression analysis was performed to study the association between the expression levels of H3K27M and H3K27me3 and the overall survival. Results The positive expression rate of H3K27M was 65.33% and the high expression rate of H3K27me3 was 58.67% in diffuse spinal glioma tissues. The expression levels of H3K27M and H3K27me3 were related to WHO grade and intracranial spread and metastasis (all P<0.05). The 2-year survival rate of the H3K27M positive group was lower than that of the H3K27M negative group (51.6% vs 78.9%, χ2=7.954, P=0.005), and the 2-year survival rate of the H3K27me3 high expression group was lower than that of the H3K27me3 low expression group (48.4% vs 74.2%, χ2=8.130, P=0.004). After adjusted for potential covariates by multivariable Cox regression, the H3K27M positive expression group had a risk of death 67% higher than that of the H3K27M negative expression group (HR=1.67, 95%CI: 1.06-2.64, P=0.026), and the H3K27me3 high expression group had a risk of death 84% higher than that of the H3K27me3 low expression group (HR=1.84, 95%CI: 1.05-3.22, P=0.032). Conclusions The H3K27M is positively expressed and H3K27me3 is highly expressed in diffuse spinal glioma tissues, both of which are associated with poor prognosis.

     Objective To observe the efficacy and safety of Callispheres drug-eluting bead transarterial chemoembolization (DEB-TACE) on the unresectable colorectal cancer liver metastases with systemic chemotherapy failure. Methods The clinical data of 42 patients with unresectable colorectal cancer liver metastasis who failed at least second-line systemic chemotherapy and admitted to Linyi Cancer Hospital of Shandong province from June 2018 to June 2020 were retrospectively analyzed. All patients received TACE with 100-300 μm Callispheres microspheres loaded with irinotecan. The tumor response was evaluated after DEB-TACE, the overall survival (OS), progression-free survival (PFS), adverse reactions during treatment and the factors affecting the OS were analyzed. Results At 2, 4 and 6 months after DEB-TACE, the objective response rate (ORR) were 83.3%, 81.0% and 71.4%, respectively; the disease control rate (DCR) were 100.0%, 90.5% and 80.9%, respectively; the median PFS was 11.0 months and the median OS was 16.0 months. The main adverse reactions were fever, pain, nausea and vomiting, which occurred mainly in grade 1-2 patients, and no serious complications such as ectopic embolism occurred. Multivariable Cox regression analysis showed that simultaneous liver metastasis (HR=0.446, 95%CI: 0.279-0.780, P=0.002)  and extrahepatic metastasis  (HR=0.311, 95%CI:0.134-0.968, P=0.007) were independent risk factors affecting OS. Conclusions DEB-TACE has certain clinical efficacy and manageable adverse reactions in the treatment of patients in unresectable colorectal cancer liver metastases with systemic chemotherapy failure, while the prognosis of patients with simultaneous liver metastasis or extrahepatic metastasis is poor.

    Construction of prognostic model for endometrial cancer based on immunerelated genes

    Objective To establish a prognostic model of immune-related genes in endometrial cancer to provide the reference for the immunotherapy of endometrial cancer.  Methods Endometrial cancer transcriptome data and clinical information were downloaded from TCGA database, and the immune-related genes were acquired from IMMPORT database. Subsequently, the differential analysis and functional enrichment analysis were used to evaluate the function of differential expression immune-related genes. The prognostic assessment models was constructed in training sets using univariable, multivariable Cox regression and least absolute shrinkage and selection operator (LASSO). The receiver operating characteristic (ROC) curve, calibration plot, decision curve analysis (DCA) curve were used to evaluate the performance of models. Results The immune-related gene expression profiles for 504 cases of endometrial cancer and 22 cases of adjacent tumor tissues were analyzed, and 394 differentially expressed genes were obtained, including 111 up-regulated genes and 283 down-regulated genes. Functional enrichment analysis showed that differential genes were mainly involved in biological processes such as immune response, signal transduction and immune response. A total of 31 prognosis-related genes were identified in the training cohort by the univariable Cox regression analysis. The predictive model of 6 immune-related genes (ANGPT4, HGF, PLA2G2A, SPAG11B, SST, VGF) were constructed by LASSO-Cox regression. The area under the ROC curve of the model in the training cohort were 0.986, 0.846 and 0.849 at 1, 3 and 5 years, respectively; the area under the curve of the model at 1, 3 and 5 years were 0.667, 0.629 and 0.693, respectively, in the validation cohort; and the area under the curve of the model at 1, 3 and 5 years were 0.798, 0.735 and 0.770, respectively, in total cohort. Conclusions The prognostic model is developed based on 6 immune-related genes, and can be effectively used to predict the prognosis of patients with endometrial cancer, providing a reference for immunotherapy.

    Objective To evaluate the impact of the prognostic nutritional index (PNI) on the prognosis of the breast cancer patients receiving neoadjuvant chemotherapy (NAC). Methods A single-center, retrospective cohort study was performed on the breast cancer patients, who underwent NAC and surgical resection in Internal Medicine-Oncology Beijing Shijitan Hospital Affiliated to Capital Medical University from January 2008 to December 2017, were selected as the research subjects, and the follow-up was as of December 2020. The main study endpoint was the disease-free survival. The cut-off values of PNI before NAC (pre-PNI), PNI after NAC (post-PNI), and PNI before and after NAC (ΔPNI) were calculated by the time-dependent ROC curve. The association between PNI and disease-free survival was analyzed by Cox proportional hazard regression. Results A total of 131 patients met the criteria. The post-PNI was lower than pre-PNI (46.4±4.5 vs 52.5±4.2, P<0.001). The cut-off value of ΔPNI was 5.26. The disease-free survival rate of the high ΔPNI group was significantly lower than that of the low ΔPNI group (P<0.001). Further multivariable Cox proportional hazard regression showed that ΔPNI, pre-NAC clinical stage, and HER-2 were independent influencing factors of the disease-free survival (all P<0.05). Conclusions The decrease in PNI during NAC indicates a poor prognosis. Maintaining a nutritional status during NAC may yield better treatment results for the breast cancer patients.

    Application of different hepatic blood flow occlusion methods in liver cancer resection

     Objective To investigate the value of the selective Glisson's pedicle transection method and the first portal occlusion (Pringle) method in liver cancer resection. Methods The clinical data of 93 patients with primary liver cancer in the Third People's Hospital of Hechi from May 2012 to May 2021 were retrospectively analyzed. The patients were divided into two groups according to the adopted intraoperative hepatic blood flow occlusion method. The control group received the Pringle method (n=46), and the treatment group received selective Glisson's pedicle transection method (n=47). The evaluation indexes included intraoperative blood loss, duration of operation, postoperative albumin input, alanine aminotransferase (ALT), total bilirubin (TBIL), anal exhaust time and hospital stay, and so on. Results There were no significant differences in duration of operation and blood loss between two groups (all P>0.05). The postoperative albumin input, ALT and TBIL in the treatment group were lower than those in the control group on the 1st, 3rd, 5th, 7th and 10th day after surgery (all P<0.05). The time of postoperative anal exhaust and hospitalization in the treatment group were shorter than those in the control group (all P<0.05). The main complications in two groups were abdominal or pleural effusion, lung infection and atelectasis, but there was no statistically significant in the incidence (all P>0.05). Conclusions In liver cancer resection, the selective Glisson's pedicle transection method is comparable to Pringle method, but with less postoperative liver function damage, faster gastrointestinal function recovery, shorter hospital stay and no increase in postoperative complications. Further randomized controlled trials are necessary.

    Effect of electroacupuncture and medicine combination on gastrointestinal function in patients with colorectal cancer after laparoscopic surgery based on the concept of enhanced recovery after surgery

    Objective To investigate the efficacy and safety of electroacupuncture (EA) combined with Evodia hot compresson on the recovery of gastrointestinal function in patients with colorectal cancer after laparoscopic surgery based on the enhanced recovery after surgery (ERAS). Methods A total of 60 colorectal cancer patients who were hospitalized in the General Surgery Department of Guizhou Provincial People's Hospital from October 2019 to December 2020 were selected and randomly divided into observation group and control group with 30 cases each. The control group was treated with the concept of ERAS during the perioperative period, and the observation group was additionally treated with EA at Zusanli and Shangjuxu acupuncture points plus Evodia hot compress Shenque acupuncture point compared to the control group. The first exhaust time, first defecation time, restore semi-liquid diet time, the time of hospitalization after operation, the abdominal pain score, the abdominal distention score, the inflammatory indicators and postoperative complications between two groups were observed. Results Compared to the control group, the observation group has significantly shorter first flatus time, first defecation time and recover semi-liquid diet time (all P<0.05), higher postoperative abdominal pain relief degree (P<0.05), lower related inflammatory indicators on the fifth day (all P<0.05), and less postoperative hospitalization days (P<0.05). During the treatment period, there were no serious complications and adverse reactions such as anastomotic leakage in the two groups. Conclusions With the concept of enhanced recovery after surgery, the electroacupuncture combined with Evodia hot compress can promote the recovery of gastrointestinal function in patients with colorectal cancer after laparoscopic surgery, and alleviate the symptoms of abdominal pain and inflammatory stress responses, without increasing the incidence of postoperative complications. It can be an effective means of postoperative gastrointestinal function recovery.

    Objective To investigate the features of contrast-enhanced ultrasound (CEUS) and ultramicrovascular imaging in breast sclerosing adenosis (SA). Methods The CEUS and ultramicrovascular imaging of 15 SA patients and 40 cases of infiltrating ductal carcinoma (IDC) patients admitted to Guangxi Medical University Cancer Hospital from December 2017 to December 2021 were analyzed retrospectively, and the CEUS and ultramicrovascular imaging features of SA were analyzed combined with pathological features. Results In the qualitative features of CEUS, there were statistically significant differences between SA and IDC in enhancement intensity, lesion edge, perfusion defects, lesion scope and ultramicrovascular imaging architecture (all P<0.05). Among quantitative parameters, there were statistically significant differences between SA and IDC in peak intensity, curve slope, area under the curve, inflow area and outflow area (all P<0.05). Conclusions The certain characteristics in CEUS and ultramicrovascular imaging of breast SA are significantly different from IDC, and a comprehensive understanding of these features helps improve the diagnostic accuracy of breast sclerosing adenosis.

    Predictive value of preoperative CT signs combined with α?fetoprotein in early recurrence of solitary hepatocellular carcinoma

    Objective To investigate the predictive value of preoperative CT signs combined with α-fetoprotein (AFP) in early postoperative recurrence of solitary hepatocellular carcinoma (HCC). Methods The patients with solitary HCC admitted to Guangxi Medical University Cancer Hospital from June 2015 to May 2017 were selected as the study subjects, and clinical data and CT signs were collected. The early recurrence factors were analyzed by logistic regression and the predictive model was established. The efficacy of preoperative CT signs in predicting early recurrence was evaluated by the receiver operating characteristic (ROC) curve. Results Among 194 patients with solitary HCC, 104 patients had early postoperative recurrence. The high AFP level (OR=1.900, 95%CI: 1.008-3.581, P=0.047), non-smooth tumor margins (OR=2.095, 95%CI: 1.078-4.072, P=0.029) and intratumoral vessels in arterial phase (OR=2.374, 95%CI: 1.186-4.754, P=0.015) were independent predictors of early recurrence of solitary HCC. The area under the ROC curve of preoperative CT signs with non-smooth tumor margins and intratumoral vessels combined with AFP in predicting early recurrence of solitary HCC was 0.725 (95%CI: 0.656-0.786), with a sensitivity of 65.4% and a specificity of 68.9%. Conclusions Preoperative CT signs combined with AFP have certain value in predicting the early recurrence of solitary HCC.
