    Objective To analyze the epidemiological characteristics and disease burden of malignant tumors in Guangxi cancer registration areas in 2017, and to provide evidence for prevention and treatment of malignant tumors. Methods According to the tumor incidence and death data reported by 33 cancer registration areas in Guangxi in 2017, the incidence, mortality and corresponding standardized rates of Chinese population (ASR China) and world population (ASR World) were calculated to analyze the epidemiological characteristics of malignant tumors. The years of lived with disability (YLDs), years of life lost (YLLs) and disability-adjusted life years (DALYs) related to cancer were collated to assess the disease burden of malignant tumors. Results In 2017, a total of 34,105 new cases of malignant tumors were reported in Guangxi cancer registration areas, with a crude incidence of 243.67/105 , ASR China of 201.71/105 and ASR World of 196.51/105 . A total of 20,709 deaths were reported, with a crude mortality rates of 147.96/105 , ASR China of 117.81/105  and ASR World of 116.26/105 . The cancer of the lung, liver, female breast, colorectum, cervix, stomach, nasopharynx, uterus, prostate and ovary were the top ten for malignant tumors incidence, accounting for 74.0% of total cancer cases. The cancer of liver, lung, colorectum, stomach, female breast, cervix, nasopharynx, esophagus, leukemia and brain were the ten leading causes of cancer deaths, accounting for 82.0% of total cancer deaths. Cancers resulted in 301,738.2 DALYs in Guangxi in 2017, the DALYs rates were 2,155.86 DALYs per 105  persons. The top 10 cancers in DALYs rate were the cancers of liver, lung, colorectum, stomach, nasopharynx, female breast, leukemia, brain, esophagus and cervix. Conclusions In 2017, the incidence, mortality, and DALYs rates of malignant tumors in Guangxi were all at a high level. The lung cancer, liver cancer, colorectum cancer, nasopharynx cancer, female breast cancer and cervix cancer are the major malignant tumor for prevention and treatment in Guangxi.

    Objective To investigate the role and underlying mechanism of glucose-6-phosphate dehydrogenase (G6PD) in cisplatin resistance of ovarian cancer cells. Methods The cisplatin-resistant ovarian cancer cell line A2780/CDDP was established by continuous induction with low concentration. The differential expressed protein G6PD in A2780 and A2780/CDDP cells were analyzed by a Label-free protein quantification method. The expression levels of G6PD was detected by Western blot and RT-qPCR. siRNA-G6PD and its negative control (siRNA-NC) were transfected into A2780/CDDP cells by siRNA technique and then the effect of G6PD gene silencing was verified by Western blot. MTT experiment was used to detect cell proliferation ability and flow cytometry was used to detect the apoptosis rate. The expression levels of reactive oxygen species (ROS), malondialdehyde (MDA), reduced glutathione (GSH) and lipid peroxides related to ferroptosis were detected by flow cytometry and microplate reader. Results Compared with A2780 cells, there were 95 up-regulated proteins and 102 down-regulated proteins in A2780/CDDP cells, among which G6PD had the most significant difference in expression. Most of these proteins were enriched in metabolic pathways, and their functions were mainly related to cellular oxidative stress response and ribose phosphoric acid metabolism pathways. Compared with A2780 cells, the expression levels of G6PD mRNA and protein in A2780/CDDP cells were significantly increased (all P<0.05), the levels of ROS, MDA and the lipid peroxides related to ferroptosis were decreased (all P<0.05), while the GSH level was increased (P=0.001). After silencing G6PD, the IC50 value of cisplatin in A2780/CDDP cells was decreased ( P=0.012), the apoptosis rate was increased (P=0.006), and the levels of ROS, MDA and the lipid peroxides related to ferroptosis were increased (all P<0.05), while the GSH level was decreased (P=0.007). Conclusions Silencing G6PD may enhance cisplatin sensitivity by inducing the ferroptosis levels in cisplatin-resistant ovarian cancer cells A2780/CDDP.

    Analysis of the effect of bone mesenchymal stem cells on endometrial cancer cells based on transcriptome sequencing

    Objective To investigate the effect of bone mesenchymal stem cells (BMSCs) on the transcription level of endometrial cancer Ishikawa cells. Methods Ishikawa cells were infected with Lenti-EGFP lentivirus. The Ishikawa cells expressing the enhanced green fluorescent protein (EGFP) were cultured separately and co-cultured by contacting with BMSCs; Ishikawa cells were isolated by flow cytometry, and the differentially expressed mRNA and miRNA were analyzed and screened by transcriptome sequencing. The differential miRNA target genes was predicted through miRWalk, and then the intersection genes of the two genomes were analyzed by GO , KEGG and protein-protein interaction network (PPI) analysis. The accuracy of the sequencing was verified by qRT-PCR and Western blot. Results After co-culture of EGFP-labeled Ishikawa cells with BMSCs, 33.6% Ishikawa-EGFP positive cells and 10.5% Ishikawa-EGFP negative cells were isolated. A total of 5 928 differentially expressed mRNA, 111 differentially expressed miRNA were screened after sequencing. PPI analysis showed that the interaction occurred among the proteins expressed by the intersection genes, and the core nodes included CDKN1A, JAK1, COL1A1, VCAN, etc. The miRNA-mRNA network diagram showed that a potential targeting relationship existed between CDKN1A and hsa-let-7e-5p. GO and KEGG analysis showed that the intersection genes were involved in extracellular matrix organization, cell adhesion molecule binding, and mainly enriched in PI3K-Akt, focal adhesion, MAPK, EGFR tyrosine kinase inhibitor resistance and other signaling pathways. qRT-PCR and Western blot showed that the expression of CDKN1A was up-regulated and the expression of hsa-let-7e-5p was down-regulated in Ishikawa-EGFP cells after co-culture. Conclusions The BMSCs may regulate the local tumor microenvironment through the interaction between cells and promote the occurrence and development of the endometrial cancer.

    Objective To construct the apatinib@zeolitic imidazolate framework-8 (A@ZIF-8) nanoparticles, and to investigate the effect of A@ZIF-8 on the proliferation and apoptosis of mouse breast cancer 4T1 cells. Methods The zeolitic imidazolate framework-8 (ZIF-8) was prepared by a physical agitation method, the apatinib was loaded into ZIF-8 by the one-step method to construct the A@ZIF-8. The particle size and morphological characteristics of ZIF-8 and A@ZIF-8 were observed by transmission electron microscope (TEM) and scanning electron microscope (SEM), the Zeta potentials were measured by laser particle size analyzer. CCK-8 assay and flow cytometry were used to detect the effects of A@ZIF-8 on the proliferation and apoptosis of 4T1 cells. One mouse was randomly selected from 13 Balb/c female mice for A@ZIF-8 in vitro hemolysis experiment. The remaining mice were randomly divided into the control group (n=6) and the A@ZIF-8 group (n=6), and sacrificed on the 14th day after the first tail vein injection. Blood was collected for blood routine and blood biochemical index detection, and HE staining was used to observe the changes of organs, including heart, liver, spleen, lung, kidney and others, to evaluate the biological safety of A@ZIF-8. Results Both TEM and SEM showed that compared with the control group, the particle size of ZIF-8 increased after loaded with apatinib and the Zeta potential increased significantly (all P<0.001), indicating that A@ZIF-8 was successfully constructed. CCK-8 assay showed that the inhibitory effect of A@ZIF-8 on 4T1 cell proliferation increased with the increase of its concentration, IC50=27.69 μg/mL. Flow cytometry showed that the apoptosis rate of the A@ZIF-8 group was higher than that of control group (P<0.001). Hemolysis test showed that the A@ZIF-8 was not prone to hemolysis in mice. After injection of A@ZIF-8 solution, the blood routine, liver function, kidney function and other indicators of mice were within the normal range, and no cell structure damage was found in HE staining of mouse vital organs under microscope. Conclusions The A@ZIF-8 constructed in this study can inhibit the proliferation of breast cancer 4T1 cells and promote cell apoptosis, with good biosafety, which is expected to be used for the treatment of breast cancer in vivo.

     Expression of LINC01215 in colorectal cancer tissues and its clinical significance

     Objective To investigate the expression of LINC01215 in colorectal cancer (CRC) tissues and its clinical significance. Methods The expression of LINC01215 in CRC tissues and adjacent normal tissues, and its relationship with prognosis were analyzed by TCGA database. The expression levels of LINC01215 in cancer tissue and adjacent normal tissue samples of 137 CRC patients diagnosed and treated in the Second Affiliated Hospital of Guangzhou Medical University from March 2009 to January 2012 were detected by RT-qPCR and the relationship between these expressions and clinicopathological characteristics was analyzed. The LINC01215 target genes were obtained from the MEM website and GO annotation and KEGG analysis were performed. According to the result of functional analysis, single-sample gene set enrichment analysis (ssGSEA) and drug sensitivity analysis were performed on LINC01215. Results TCGA database analysis showed that the expression of LINC01215 in CRC tissues was lower than that of adjacent normal tissues (P<0.001). Clinical specimens verification revealed that the expression of LINC01215 in CRC tissues was lower than that of adjacent normal tissues (P=0.001), and correlated with the degree of tissue differentiation (P=0.037). TCGA database analysis showed that the median overall survival of the high LINC01215 expression group was significantly higher than that of the low LINC01215 expression group [96 months (95%CI: 80-113 months) vs 82 months (95%CI: 70-95 months), P=0.002]. Low LINC01215 expression was a risk factor for poor prognosis of CRC (HR=1.69, 95%CI: 1.13-2.51, P=0.010). GO and KEGG analysis showed that the target genes related to LINC01215 was mainly involved in protein binding, T cell receptor signaling pathway and other pathways. ssGSEA analysis found that LINC01215 was associated with 28 immune cells (all P<0.05). Drug sensitivity analysis showed that LINC01215 was related to the drug sensitivity of Chelerythrine, Sapacitabine, Hydroxyurea and Fenretinide (all P<0.05). Conclusions LINC01215 is lowly expressed in CRC and associated with poor prognosis. It may affect the occurrence and development of CRC by participating in the regulation of  T cell receptor signaling pathway and other pathways.

    Application of cone-beam breast CT volume measurement in prosthetic breast reconstruction after nipple-sparing mastectomy in breast cancer

    Objective To investigate the application value of cone-beam breast computed tomography (CBBCT) in prosthetic breast reconstruction after nipple-sparing mastectomy (NSM). Methods A total of 52 breast cancer patients who received prosthetic breast reconstruction after NSM at Guangxi Medical University Cancer Hospital from July 1, 2020 to January 31, 2022 were selected as the subjects. Spearman rank correlation was used to analyze the correlation between the measured volumes of the affected breast CBBCT and the contralateral breast CBBCT and the implanted prosthesis volume obtained by the clinical manual linear measurement method. A simple linear regression equation was established to predict the prosthesis volume calculated by CBBCT, and to compare it with the volume of implanted prosthesis. The Harris beauty score was used to evaluate the cosmetic effect 3 months after surgery. Results All patients completed breast prosthesis reconstruction without prosthesis rupture, prosthesis displacement and other complications. The measured volumes of the affected breast CBBCT and the contralateral breast CBBCT were correlated with the implanted prosthesis volume (rs=0.73, 0.81). A linear prediction model y=0.001x+4.897 (R2=0.7, P<0.001) was constructed based on the measured volume of the contralateral breast CBBCT. The clinical validation showed that there was no significant difference between the model predicted volume and the implanted volume of prosthesis (t=0.03, P=0.98), and the mean absolute error value was only 0.1 cm3. The Harris beauty score showed that the cosmetic effect was excellent in 4 cases and good in 6 cases 3 months after operation. Conclusions CBBCT can effectively predict the volume of implanted prosthesis in breast reconstruction surgery after NSM.

    A prospective study of the effect of lymph node dissection on the prognosis of patients with early ovarian cancer

    Objective To evaluate the effect of lymph node dissection on the prognosis of patients with early epithelial ovarian cancer (EOC). Methods The patients with stage Ⅰ-Ⅱ EOC in our hospital from January 2008 to December 2018 were retrospectively recruited. According to whether lymph node dissection was performed, the patients were divided into an operation group without lymph node dissection (No-LND group) and a lymph node dissection group (LND group). The demographic characteristics, clinicopathological information and perioperative adverse events of the two groups were collected, and Cox proportional hazard regression was used for survival analysis. Results A total of 279 patients with EOC were enrolled, 56 cases in the No-LND group and 223 in the LND group. The median number of lymph nodes removed in the LND group was 25, including 21 pelvic lymph nodes and 4 para-aortic lymph nodes. The 5-year PFS rates of the LND group and No-LND group were 85.9% and 81.6%, respectively, and the 10-year PFS rates were 81.3% and 72.9%, respectively, with a statistically significant difference (P=0.013). The 5-year OS rates of the LND group and No-LND group were 93.8% and 88.7%, respectively, and the 10-year OS rates were 86.7% and 82.6%, respectively, with a statistically significant difference (P=0.042). Multivariable Cox regression analysis showed that lymph node dissection was a protective factor for prognosis (HR=0.89, 95%CI: 0.79-0.97, P=0.041), but the median operation time of the LND group was longer than that of No-LND group (220 min vs 155 min,  P<0.001), and the incidence of lymphatic cysts at discharge was also higher in the LND group (31.8% vs 0, P<0.001). Conclusions Lymph node dissection can improve the prognosis of patients with early EOC, but it may increase the incidence of perioperative adverse events. Therefore, the therapeutic value of lymph node dissection in the patients with early EOC still needs to be further verified by large-scale, prospective randomized controlled trials.

    Analysis of ultrasonic features of pure and mixed mucinous breast carcinoma

     Objective To analyze the two-dimensional ultrasound (2DUS) and color Doppler flow imaging (CDFI) characteristics of pure mucinous breast carcinoma (PMBC) and mixed mucinous breast carcinoma (MMBC). Methods The 2DUS and CDFI images of 49 patients with mucinous breast carcinoma (MBC), who were diagnosed by surgical pathology in the First Affiliated Hospital of Guangxi Medical University from January 2013 to December 2020, were retrospectively analyzed. The ultrasonic differences between PMBC and MMBC in shape, margin, echo pattern, posterior acoustic feature, calcification, blood flow grade and BI-RADS category were compared. Results Among 49 cases of MBC lesions, there were 1 case of BI-RADS category 3, 4 cases of BI-RADS category 4a, 15 cases of BI-RADS category 4b, 11 cases of BI-RADS category 4c, and 18 cases of BI-RADS category 5. The detection rate of heterogeneously mixed echoes in the PMBC group was higher than that in MMBC group (P=0.025). The detection rates of posterior echo attenuation and spiculated indistinct margin in MMBC group were higher than those in the PMBC group (P=0.008, 0.023); The proportions of blood flow grade 3 masses and BI-RADS class 5 masses in the MMBC group were higher than those in the PMBC group; the proportion of BI-RADS class 4b masses was lower than that in the PMBC group. Conclusions Compared with MMBC, PMBC is more easily benign ultrasound features. Comprehensive analysis of the margin, echo pattern, posterior features and CDFI findings can help to distinguish PMBC from MMBC, thereby improving the ultrasound diagnostic accuracy of MBC.

    Application value of internet-based continuous management for nasopharyngeal carcinoma patients with symptoms 5 years after hospital discharge

    Objective To investigate the effect of the internet-based continuous management for the nasopharyngeal carcinoma patients with symptoms 5 years after hospital discharge. Methods Patients with nasopharyngeal carcinoma who completed the treatment in the Radiotherapy Department of the First Affiliated Hospital of Guangxi Medical University from September 2014 to March 2015 and had been discharged for 5 years were selected as the research objects. The patients were randomly divided into the control group and the intervention group. The control group took routine measures, and the intervention group adopted "Internet +" continuous management on the basis of the control group. The symptom distress, fear of disease progression and quality of life scores were compared between the two groups. Results After 3 and 6 months of intervention, the scores of symptom distress and the fear of disease progression in the intervention group were lower than those in the control group (all P<0.05), but the scores of overall life quality and functional areas were higher than those of the control group (all P<0.05), and the scores of fatigue, sleep, shortness of breath and poor appetite were lower than those of the control group (all P<0.05). Conclusions The internet-based continuous management can reduce the symptom burden, alleviate fear and improve the quality of life for the patients with nasopharyngeal carcinoma who have been discharged for 5 years.
