人肝癌细胞系中CD90+细胞的生物学特性

Biological Characteristics of CD90+ Cells in Human Hepatocellular Carcinoma Cell Lines

摘要: 目的 筛选肝癌细胞系中CD90 (Thy-1)阳性表达的细胞,初步探讨CD90+细胞亚群的干细胞特性。方法 利用流式细胞分选术筛选5种肝癌细胞株(HepG2、SMMC-7721、Bel-7404、Bel-7403及SK-Hep-1)中CD90 (Thy-1)阳性表达的细胞,以平板克隆形成实验观察其增殖能力,CCK-8法观察CD90+细胞的增殖能力和顺铂对CD90+细胞的抑制率。结果 肝癌细胞株Sk-Hep-1中有66.02%的细胞CD90呈阳性表达。平板克隆形成实验显示CD90+细胞克隆形成率为(18.60±2.95)%,显著高于CD90-细胞及未分选细胞,差异有统计学意义(P<0.05)。体外实验显示培养4 d后CD90+细胞的增殖较CD90-及未分选细胞明显加快;CD90+细胞在相同浓度的顺铂作用下其抑制率较CD90-细胞和未分选细胞明显降低,三者的抑制率分别为14.8%、29.7%和24.0%,差异有统计学意义(P<0.05)。结论 肝癌细胞中的CD90+细胞是具有肿瘤干细胞特性的细胞亚群,CD90可能是肝癌干细胞候选的分子标志物。

Abstract: Objective To isolate CD90+ (Thy-1+) cells from human hepatocellular carcinoma (HCC) cell lines and to study their stem cell properties. Methods Fluorescence-activated cell sorting was used to detect CD90 expression in five HCC cell lines:HepG2,SMMC-7721,Bel-7404,Bel-7403 and SK-Hep-1.The tablet clone formation test was used to measure the colony-forming ability of the sorted CD90+ cells,and the CCK-8 assay was used to analyze their proliferation ability and their sensitivity to cisplatin. Results CD90 expression was detected in 66.02% of Sk-Hep-1 cells.The pelletizing rate of these CD90+ cells was(18.6±2.95)% in the tablet colony formation assay, significantly higher than that of CD90- and unsorted cells(P<0.05). Proliferation after 4 days in culture was significantly greater for CD90+ cells than for CD90- and unsorted cells.Cisplatin inhibited growth of CD90+ cells by 14.8%,significantly less than it inhibited the growth of CD90- cells (29.7%,P<0.05) and unsorted cells (24.0%,P<0.05). Conclusion CD90+ cells are a subpopulation with tumor stem cell properties.CD90 may be a candidate biomarker for HCC stem cells.
