lncRNA SH3PXD2A-AS1在结肠癌组织中的表达及其临床意义

 Expression and clinical significance of lncRNA SH3PXD2A-AS1 in colon cancer tissues

摘要： 目的 探讨长链非编码RNA（lncRNA） SH3PXD2A-AS1在结肠癌组织中的表达及其临床意义。方法 收集于广西医科大学附属肿瘤医院手术切除的40例结肠癌患者的癌组织及相应癌旁正常组织。采用RT-PCR法检测lncRNA SH3PXD2A-AS1的表达水平。下载TCGA数据库中的结肠癌数据，验证lncRNA SH3PXD2A-AS表达水平与结肠癌临床病理特征及预后的关系。结果 lncRNA SH3PXD2A-AS1在结肠癌组织中的表达水平高于癌旁正常组织（6.53±1.62 vs 4.37±0.96，t=3.445，P=0.002），其在M1期患者中的表达水平高于M0期，在Ⅲ~Ⅳ期患者中患者中的表达水平亦高于Ⅰ~Ⅱ期 （P<0.05），但与患者总生存期无关（χ2=0.326，P=0.586）。TCGA数据库验证结果显示，lncRNA SH3PXD2A-AS1在结肠癌组织中呈高表达，但与患者的总生存期无关（P>0.05）。结论 lncRNA SH3PXD2A-AS1在结肠癌组织中呈高表达，且与肿瘤远处转移有关，可能是判断肿瘤进展程度的指标。

Abstract: Objective To investigate the expression of long-chain non-coding RNA（lncRNA） SH3PXD2A-AS1 in colon cancer tissues and its clinical significance. Methods The tumor tissues and corresponding normal tissues were collected from 40 cases of surgical resection at Guangxi Medical University Cancer Hospital. RT-PCR was used to detect the expression level of lncRNA SH3PXD2A-AS1. Colon cancer data from the TCGA database were downloaded and used to verify the relationship between the expression level of lncRNA SH3PXD2A-AS1 and the clinicopathological features and prognosis of colon cancer. Results The expression level of lncRNA SH3PXD2A-AS1 in colon cancer tissues was higher than that in normal adjacent tissues（6.53±1.62 vs 4.37±0.96，t=3.445，P=0.002）. The expression level in the M1 state was higher than that in M0 state，and the expression level of stage Ⅲ-Ⅳ was also higher than that of stage Ⅰ-Ⅱ（P<0.05），though it was not related to the overall survival（χ2=0.326，P=0.568）. Results from the TCGA database showed that lncRNA SH3PXD2A-AS1 was highly expressed in colon cancer tissues，though it was not related to the overall survival（P>0.05）. Conclusions lncRNA SH3PXD2A-AS1 is highly expressed in colon cancer tissues and related to distant metastasis of the tumor，which may become an indicator of the degree of tumor progression.
