    Effects of taurine on tumor growth in a nude mouse breast cancer model

    Objective To study the effect of taurine on the humanized nude mouse model of triple-negative breast cancer. Methods Five-week-old SPF level nude mice(n=10) were inoculated with MDA-MB-231 breast cancer cells and randomly divided into one group that received normal drinking water and another group that received water containing 5% taurine(n=5 animals per group). Tumor volume and weight were examined after treatment, and Western blotting was used to assess expression of estrogen receptor (ER), progesterone receptor(PR),human epidermal growth factor receptor 2 (HER-2)and guanosine triphosphatase-activating protein p21ras. Results All mice survived until the end of the experiment,and all tumors grew. Compared to control mice, mice given taurine had tumors with 41.6% smaller weight and volume(P<0.05),134% higher expression of ER(P<0.05), 47.5% higher expression of PR (P<0.05),67% lower expression of HER-2(P<0.05)and 62.2% lower expression of p21ras(P<0.01). Conclusion Taurine inhibits tumor growth by up-regulating expression of ER and PR and down-regulating expression of HER-2 and p21ras.

    Objective To investigate the correlation between polymorphism at rs2298141 in the integrin β1(ITGβ1)gene and susceptibility of families in Guangxi to hepatocellular carcinoma(HCC). Methods In this study,20 HCC family groups comprising 79 members served as cases,and 10 normal family groups comprising 40 members served as controls. Time-of-flight mass spectrometry was used to determine genotype frequencies at rs2298141,and possible correlations between genotype and HCC risk were explored using non-conditional logistic regression. Results AA,AG and GG genotypes were detected at rs2298141,and their frequencies followed Hardy-Weinberg equilibrium. Frequencies of alleles A and G at rs2298141 were similar among controls,core individuals in HCC families and HCC patients (allele A:63.75%,61.02%,70%;allele G:36.25%,38.98%,30%;P>0.05). Relative to risk of HCC in individuals with allele A,risk of HCC in controls with allele G was 0.75(95%CI 0.33-1.7,P=0.50). Relative to risk of HCC in families with allele A,risk of HCC in HCC families with allele G was 0.67(95%CI 0.31-1.45,P=0.31). AA,AG,and GG genotypes occurred at respective frequencies of 40%,42.73%,and 50% among controls;47.5%,37.29%,and 40% among core individuals in HCC families;and 12.5%,20.34%,and 10%among HCC patients(P>0.05). The corresponding frequencies of genotypes AA,AG and GG in the three groups was 40.00%,47.50% and 12.50%;50.00%,42.73%,37.29% and 20.34%;and 50%,40% and 10%. Allele distributions did not differ significantly between cases and controls. Relative to risk of HCC in individuals with the AA genotype, risk of HCC in control families with genotypes AG or GG was,respectively,0.67 (95%CI 0.22-2.1,P=0.50) and 1.05(95%CI 0.17-6.6,P=0.96), while the risk of HCC in HCC families with genotypes AG or GG was, respectively, 0.9(95%CI 0.31-2.71,P=0.86) and 2.2(95%CI 0.40-11.96,P=0.37). Conclusion The ITGβ1 rs2298141 polymorphism does not appear to correlate with susceptibility to HCC based on family clustering in Fusui County,Guangxi.

    NF-κB,COX-2 and LKB1 expression in endometrial cancer tissues and clinical significance

    Objective To analyze NF-κB,COX-2 and LKB1 expression in endometrial cancer tissues and normal controls,and assess clinical significance. Methods Immunohistochemistry was used to analyze expression of NF-κB,COX-2 and LKB1 in 60 endometrial cancer tissues,60 normal endometrial tissues from uterine myomas and 31 cases of atypical hyperplasia of endometrium. Possible relationships were explored between NF-κB,COX-2 and LKB1 expression and clinicopathological variables. Results The rate of NF-κB positivity was 63.3% for endometrial cancer tissue,41.9% for normal endometrial tissue,and 35.0% for atypical hyperplasia tissue. The corresponding results for COX-2 positivity were 70.0%,64.5% and 51.7%. The corresponding results for LKB1 positivity were 35.0%,61.3% and 93.3%. Expression of NF-κB and LKB1 varied significantly across the three types of tissue(P<0.05),but expression of COX-2 did not(P>0.05). Expression of none of the three proteins correlated with pathology type,clinical stage or lymph node metastasis;however,LKB1 expression correlated positively with myometrial invasion and histology grade(P<0.05),while NF-κB expression correlated with histology grade(P<0.05). Conclusion NF-κB is over-expressed and LKB1 under-expressed in endometrial cancer tissue. These proteins may be involved in the development and progression of endometrial cancer.

    Effects of postoperative chemoradiotherapy on locally advanced esophageal squamous cell carcinoma

    Objective  To evaluate the efficacy of chemoradiotherapy for locally advanced esophageal squamous cell carcinoma. Methods  Clinical data were retrospectively analyzed for 214 patients with esophageal cancer who underwent surgery between 2008 and 2011. Rates of local recurrence,distant metastasis and survival were compared between 121 patients who underwent both surgery and postoperative chemoradiotherapy,and 93 patients who underwent surgery alone. Results  Patients receiving postoperative chemoradiotherapy showed a significantly lower rate of local recurrence than patients who underwent surgery alone(27.38% vs 45.37%);a similar result was observed for rate of distant metastasis(31.65% vs 46.23%)(P<0.05). Similarly,the postoperative chemoradiotherapy group showed significantly higher overall survival rates at 1 year(98% vs 90%),3 years(74% vs 55%),and 5 years(52% vs 23%). All patients tolerated their treatment well. Most adverse events were grades 1 and 2;grade 3 adverse events were leucopenia,radiation esophagitis,cough,and anastomotic stenosis. Conclusions Postoperative chemoradiotherapy can provide overall survival benefit to patients with locally advanced esophageal squamous cell carcinoma.

    Relationship between subjective well-being and social support in elderly patients with prostate cancer

    Objective To study subjective well-being,social support and their relationship in elderly patients with prostate cancer. Methods A total of 200 elderly patients with prostate cancer admitted for uropoiesis surgical treatment from August 2014 to August 2015 filled out a happiness index scale,social support rating scale,and a custom-designed general questionnaire. Results The subjective well-being score of elderly patients was 7.05±3.48,and the social support score was 28.75±4.37. Subjective well-being and social support scores varied significantly with different disease progression,culture,marital status and number of hospitalizations(P<0.05). Correlations were observed among social support,subjective support,objective support,social support utilization(three dimensions),subjective well-being,total emotion index and life satisfaction(P<0.001). Total score for social support correlated with subjective well-being score(r=0.834,P<0.001). Conclusion Subjective well-being and social support scores were low for elderly patients with prostate cancer. Subjective well-being in these patients strongly correlated with social support. Health care workers and their families should pay more attention to providing social support to such patients and helping them expand their social support system in order to improve their subjective well-being and prolong their quality of life.

    Efficacy and safety of capecitabine maintenance therapy for patients with advanced gastric cancer after first-line XELOX chemotherapy

    Objective To analyze the efficacy and safety of capecitabine maintenance treatment in patients with advanced gastric cancer after successful chemotherapy through the first-line XELOX program(oxaliplatin and capecitabine). Methods Patients with advanced gastric cancer(n=84)who completed first-line XELOX chemotherapy were randomly divided into a control group(n=42)that received no further treatment or a maintenance group(n=42)that received capecitabine chemotherapy until disease progression or intolerable toxicity. Results Progression-free survival was 9.6 months in the maintenance group,significantly longer than the 6.8 months in the control group(P<0.05). Similarly,overall survival was 13.5 months in the maintenance group,significantly longer than the 11.6 months in the control group(P<0.05). The main adverse reactions were hematologic toxicity,gastrointestinal reaction,hand-foot syndrome,oral mucositis,and peripheral neurotoxicity. All these reactions improved after symptomatic treatment. No treatment-related deaths occurred. Conclusion Capecitabine maintenance treatment can prolong progression-free survival and overall survival in patients with advanced gastric cancer who have undergone the first-line XELOX program. Toxic effects of such treatment are well tolerated.

    MRI for evaluating the effects of radiotherapy combined with other interventions on local advanced cervical cancer

    Objective To assess the usefulness of MRI for evaluating radiotherapy combined with other interventions to treat local advanced cervical cancer. Methods A total of 29 patients with cervical cancer were treated with radiotherapy and other interventions. All patients underwent conventional-sequence pelvic MRI scanning and diffusion-weighted MR imaging before intervention and after the first,second and third rounds of radiotherapy. Apparent diffusion coefficient (ADC) and tumor size were determined. Based on the effects of the intervention,the patients were classified as responders or non-responders. Changes in ADC were compared between the two groups,and survival time was compared among patients with different ADC values. Results Of the 29 treated patients,20 responded and 9 did not. Conventional MRI imaging revealed low signal in T1W1 images and higher,more homogeneous signal in T2W1 images. DWI images revealed heterogeneous high signal and clear lesion borders;ADC images revealed uneven lesions of low signal. ADC values after each radiotherapy round were significantly higher in responders than in non-responders(P<0.05). Survival time was significantly longer in patients whose ADC changed by more than 35% than in those whose ADC changed by less than 35%(P=0.021). Conclusion MRI,DWI and ADC values can be used to assess the effect of combined radiotherapy and other interventions to treat cervical cancer.

    Relationship between low-dose radioactive zone volume and radiation-induced lung injury in patients with non?鄄small cell lung cancer

     Objective  To analyze the relationship between low-dose radiation dosimetry parameters and occurrence of radiation-induced lung injury in patients with non-small cell lung cancer(NSCLC). Methods  Medical records were retrospectively analyzed for 103 patients with NSCLC who received radiation therapy. Single-factor analysis was used to explore correlations of dosimetric parameters and occurrence of grade 2 or higher radiation-induced lung injury. Parameters that significantly predicted injury were assessed for their predictive ability by examining the area under the receiver operating characteristic curve(AUC). Results  Of the 103 patients,27(26.2%)experienced radiation-induced lung injury of grade 2 or higher. Dosimetric parameters showed the following AUCs for predicting such injury: double lung V5,0.714;double lung MLD,0.673;ipsilateral lung V5,0.690;ipsilateral lung V10,0.693;and healthy contralateral lung V5,0.737. Incidence of radiation-induced lung injury was significantly higher in patients with double lung V5 ≥52.22%,double lung MLD≥1 486.75 cGy,ipsilateral lung V5 ≥62.03%,healthy contralateral lung V5 ≥43.60%,or ipsilateral lung V10 ≥55.67%. Conclusion Radical radiotherapy planning should take into account the influence of low-dose radiation on risk of radiation-induced lung injury.Except common parameters,such as V20,V30 and MLD,V5 should also be considered.

    Clinical analysis of uterine cystic lymphangioma: a case report and literature review

    Objective To explore the clinical pathological features of uterine cystic lymphangioma and promote proper understanding among clinical gynecologists. Methods Clinical manifestations,auxiliary examinations,imaging outcomes,differential diagnosis and treatment of a case of uterine cystic lymphangioma were analyzed,and related literature was reviewed. Results We report the case of a 45-year-old woman hospitalized for intermittent abdominal pain and an abdominal mass. On the basis of these factors,combined with relevant auxiliary examinations,the patient was diagnosed with uterine fibroid cystic change  before the operation.The patient was treated by total hysterectomy,a cystic mass arising from the anterior uterine wall,and clear yellow fluid filled the cyst,while the interior uterine wall is smooth,the uterine cavity is not connected with the cyst. The capsule wall of the tumor was composed of simple squamous cell,lymphatic network structure was obsersed in the smooth muscle of the cystic wall,chamber fibrin content and a few lymphocytes were  also observed. D2-40 was positive,CKpan was weakly positive. The pathological diagnosis was uterus cystic lymphangioma. No special treatment was given after surgery,there was no recurrence at the follow-up of 6 months. Conclusion Uterine adenomyoma cysts are extremely rare and are characterized by abnormal vaginal bleeding and pelvic mass. No features are visible by imaging,serum tumor markers are usually not elevated,and the disease is easily misdiagnosed. Surgery is the treatment of choice,and pathology and immunohistology tests helps to confirm the diagnosis.
