 PRR11和SKA2在宫颈癌组织中的表达及其与预后的关系

Expression of PRR11 and SKA2 in cervical cancer tissues and its relationship with prognosis

摘要: 目的 探讨PRR11(priline-rich 11)和SKA2 (spindle and kinetochore associated complex subunit 2)在宫颈癌组织中的表达及其与预后的关系。方法 收集2012年1月—2015年12月于青岛大学附属烟台毓璜顶医院诊治的289例宫颈癌患者癌组织及其癌旁正常组织,采用免疫组化检测PRR11和SKA2在宫颈癌组织中的表达,并分析其与患者临床病理特征的关系,采用Spearman法分析两者表达的相关性,Kaplan-Meier分析总生存率,Cox比例风险回归模型分析预后的影响因素。结果 PRR11和SKA2在宫颈癌组织中的阳性表达率均高于癌旁正常组织(60.21% vs 16.96%,χ2=114.081,P<0.001; 69.55% vs 37.37%,χ2=60.143,P<0.001)。Spearman相关分析结果显示,PRR11与SKA2蛋白表达呈正相关(r=0.725,P<0.001)。PRR11和SKA2蛋白表达均与FIGO分期、组织分化程度及淋巴结转移有关(P<0.05)。PRR11、SKA2阳性表达患者的3年总生存率均明显低于阴性表达患者(78.16% vs 94.78%,P=0.002; 82.09% vs 90.91%,P=0.036)。FIGO分期Ⅲ~Ⅳ期、PRR11阳性表达和SKA2阳性表达是影响宫颈癌患者预后的独立危险因素(HR=1.698,95%CI1.369~2.876,P<0.001;HR=2.392,95%CI:1.294~3.596,P<0.001;HR=1.597,95%CI:1.024~2.302,P<0.001)。结论 PRR11和SKA2在宫颈癌组织中高表达且呈正相关,是影响患者预后的独立危险因素,可能是宫颈癌预后评估的有效指标。

Abstract:  Objective To investigate the expression of priline-rich 11(PRR11) and spindle and kinetochore associated complex subunit 2(SKA2) in cervical cancer tissues and its relationship with prognosis. Methods The cervical cancer tissues and adjacent normal tissues of 289 patients with cervical cancer treated in Yantai Yuhuangding Hospital affiliated to Qingdao University from January 2012 to December 2015,were collected. The expression of PRR11 and SKA2 proteins in cervical cancer tissues was detected by immunohistochemiscal staining,and their relationship with the clinicopathological characteristics of patients was analyzed. The Spearman method was used to analyze the correlation between PRR11 and SKA2 expression,Kaplan-Meier was used to analyze the overall survival rate,and Cox proportional risk regression model was used to analyze the prognostic factors. Results The positive expression rate of PRR11 and SKA2 in cervical cancer tissues were higher than those in adjacent normal tissues(60.21% vs 16.96%,χ2=114.081,P<0.001;69.55% vs 37.37%,χ2=60.143,P<0.001). The Spearman correlation analysis results showed that PRR11 was positively correlated with SKA2 protein expression(r=0.725, P<0.001). Both PRR11 and SKA2 protein expression were related to FIGO stage,tissue differentiation degree and lymph node metastasis(P<0.05). The 3-year OS of patients with PRR11 and SKA2 positive expression were significantly lower than those of patients with negative expression(78.16% vs 94.78%,P=0.002;82.09% vs 90.91%,P=0.036). FIGO stage Ⅲ-Ⅳ,PRR11 positive expression and SKA2 positive expression were the independent risk factors affecting the prognosis of cervical cancer patients(HR=1.698,95%CI:1.369-2.876,P<0.001;HR=2.392,95%CI:1.294-3.596,P<0.001;HR=1.597,95%CI:1.024-2.302,P<0.001). Conclusion PRR11 and SKA2 are highly expressed and positively correlated in cervical cancer tissues. They are independent risk factors affecting the prognosis of patients and may become effective indicators for the diagnosis and prognosis assessment of cervical cancer.
