Effect of LRP1 on the incidence of prostate cancer based on genome-wide ChinaPca data and gene expression

Abstract: Objective To investigate the effect of low density lipoprotein receptor-related protein-1 (LRP1) gene on prostate cancer (Pca). Methods Based on ChinaPca GWAS data,nine single nucleotide polymorphism(SNP) of LRP1 were given to verify the prostate cancer susceptibility. Immunohistochemical tests were performed in 12 Pca tissues,18 normal prostate tissues and 4 prostatitis tissues,respectively. LRP1 gene expression data from TCGA and GTEx databases were analyzed,and the interaction network of LRP1 genes on the String website was built. Results ChinaPca GWAS data showed that rs11172113 of LRP1 gene might be the susceptible site of Pca(P=0.083),and TC genotype was related to the risk of Pca(OR=1.267,95%CI:1.066-1.505,P=0.007). The results of immuno - histochemistry experiment and TCGA database analysis showed that there was no statistically significant difference in the expression of LRP1 in Pca tissues and normal prostate tissues(P>0.05). After integrating the data in TCGA and GTEx databases,the expression of LRP1 in Pca tissues was decreased(P<0.05). The interaction network showed that LRP1 was associated with the PLAT,PDGFB and PDGFRB  genes  which related to the prostate cancer pathway. Conclusions rs11172113 of LRP1 may be a genetic predisposition to prostate cancer. The low expression of LRP1 gene in prostate cancer. LRP1 may regulate the prostate cancer-related pathways by regulating the expression of PLAT,PDGFB and PDGFRB genes and affect the incidence of prostate cancer.
