    The implications of hOGG1 expression in hepatocellular carcinoma and cirrhosis tissues

    Aim:To explore the expression and its clinicopathological implications of human 8-oxoGuanine DNA glycosylase-1 (hOGG1) in hepatocellular carcinoma(HCC) and cirrhotic tissues.Methods:hOGG1 expression was detected by immuneohistochemical EnVision method on liver tissues of non-HCC controls(n =41),cirrhotic(n =51) and HCC tissues(n =99 ).Results: There were three types of hOGG1 positive staining in HCCs:nuclear,cytoplasmic and mixed.The expression levels of hOGG1 in nucleus was significantly higher in HCCs than that in cirrhosis and controls and significantly higher in cirrhosis than that in controls(P<0.01).The expression of hOGG1 was not associated with the clinicopathological factors in HCCs(P>0.05 ).Multiplicity analysis indicated that hOGG1 expression in nucleus was related to ALT(OR = 1.022,P = 0.041),hOGG1 expression in cytoplasm was related to T bili(OR = 1.155,.P = 0.018).Conclusion:During hepatocarcinogenesis,the expression pattern of hOGG1 in liver cell nucleus might reflect the status of oxidative stress in each stage thus has etiology implications.

    An experimental study for mAFP gene-modified DC tumor vaccine on transplanted liver cancer

    Objective To investigate the inhibitory effect of pAdBM5-mAFP-DC tumor vaccine on the transplanted liver cancer in mice.Methods 40 C57BL/6J mice were randomly divided into group A,B,C,D and E(8 mice in each group).Group A was immunized with mAFP gene-modified DC(pAdBM5-mAFP-DC);Group B was immunized with simple pAdBM5 plasmid; Group C was immunized with recombinant pAdBM5-mAFP plasmid;Group D was immunized with simple DC;Group E was injected with PBS.Each mouse in group A and D was immunized with 5×10~5 cells,in group B and C was immunized with 10.0μg plasmid DNA,in group E was injected with 0.1ml PBS.They were immunized once a week for a consecutive 4 weeks.At the third week,all the mice were transplanted with Hepa1-6 cells.All mice were killed 14 days after Hepa1-6 immunization.Results There was no tumorigenesis could be observed in 3 of 8 mice in group A.The average tumor weight of group A was far more lighter than that in the other groups,with the statistical significance(P<0.05 or 0.01).The tumor-inhibited rate of group A,B,C and D were 68.31%,13.23%,52.1%and 38.54%,respectively.The tumor-inhibited rate for group A was significantly higher than that in the other groups(P<0.05 or 0.01).Conclusion mAFP gene-modified DC tumor vaccine could induce immunoprotection and reduce the risk of tumorigenesis.

    Objective To investigate the cytolytic effect against liver cancer cells of hIL-2 gene transfected cytokine induced killer cells(CIK) combined with dendritic cells(DC).Methods Peripheral blood mononuclear cells(PBMC) were isolated from healthy adult donors by density gradient centrifugation.After 2 hours incubation,CIK was generated from non-adherent cells by anti-CD3McAb,IL-2,IL-1 and IFN-γ.DC was induced from adherent cells by rhIL-4 and rhGM-CSF.Recombinant adenovirus vector pAdBM5-GFP-IL-2 infected CIK at 100 MOI generated IL-2-CIK.DC were pulsed with HepG2 lysate at the ration of 1:3 and generate lysate-DC.The effective cells were lysate-DC-IL2-CIK,lysate-DC-CIK,CIK,DC-CIK and IL2-CIK respectively The target cells were HepG2,K562 and H7404 tumor cell line.The ratio of effective cells and target cells(E/T) were 10:1,20:1 and 40:1 respectively.The cytotoxicity against HepG2,K562 and H7404 was detected with MTT assays. Results There were stronger cytolytic effects against HepG2 for effective cells(Lysate-DC-IL2-CIK) than those effective cells against K562,H7404 and HepG2 in the control group at different E/T ratios.Its cytolytic effects enhanced with the rising of E/T ratio and reached climax when the E/T ratio was 1:40.Conclusions hIL-2 transfected CIK combined with hepatocellular carcinoma antigen pulsed DC has more effective cytolytic effect.

    Objective Multidrug resistance is one of the main causes of treatment failure of chemotherapy.Reversal agents may be used clinically.This study was to evaluate the reverse effect of microwave hyperthermia on drug-resistance of MCF-7/ ADM to adriamycinum(ADM) cells,and to explore its mechanisms.Methods Breast cell line MCF-7 and its ADM- resistance cell line MCF-7/ADM were treated with ADM and microwave hyperthermia.Reverse effect of microwavehyperthermia on drug-resistance of MCF-7/ADM to ADM was tested by MTT assay.The levels of cellular multidrug resistance related protein(MRP) and P-glycoprotein(P-gp) were detected by Real-Time PCR.The intracellular concentration of ADM was measured by high performance liquid chromatography.Results The resistance index of MCF-7/ADM cells to ADM was 45.5.After microwave hyperthermia, the 50%inhibitory concentration(IC_(50)) of ADM to MCF-7/ADM cells reduced by 2.64 folds from(60.56±2.38)ug/ ml to(22.86±2.76)ug/ml.Protein levels of MRP and P-gp were higher in MCF-7/ADM cells than that in MCF-7 cells,and they were markedly decreased in MCF-7/ADM cells after microwave hyperthermia.The concentration of ADM in MCF-7/ADM cells was significantly increased after microwave hyperthermia.Conclusion Microwave hyperthermia may reverse drug-resistance of MCF-7/ADM cells to ADM through down-regulating MRP and P-gp and increasing intracellular cumulation of ADM.

    Objective To investigate the relationship between serial drug sensitivity or resistance to liver cancer cells and peripheral leucocytes,lymphocytes and granulocytes.Methods The sensitivity or resistance of 10 kinds of anticarcinogen for peripheral leucocytes,lymphocytes,granulocytes and the tumor cells from 31 cases of liver cancer were tested by the MTT assay. Results Except HCPT,the inhibition rates of the tested drugs in the cancer cells were apparently higher than that in the leucocytes, lymphocytes and granulocytes.The sensitivity of the top 5 anticarcinogen in cancer cells,leucocytes and lymphocytes were approximately.The correlativity between the inhibition rates of arbitrary two sets cells were all significant(P<0.01). Conclusion It was feasible to detect the sensitivity or resistance of peripheral leucocytes and lymphocytes of liver cancer patients. It could provid practical and valuable reference to optimize the individual treatment in liver cancer chemotherapy.

    Objective To investigate the correlation between serum decoy receptor 3(DcR3) and TNM staging in gastric carcinoma (GC).Methods Serum DcR3 levels were measured by using enzyme linked immunosorbent assay(ELISA) in 34 patients with GC,18 patients with nonmalignant conditions and 28 healthy individuals.Results The levels of serum DcR3 in GC were significantly higher than that in the control group(P<0.01) and the nonmalignant conditions(P<0.01),respectively. Serum DcR3 levels in GC in TNM stageⅢandⅣwere obviously higher than that in stageⅠandⅡ(P<0.01).In addition, patients with both lymph node metastasis and systemic metastasis showed significantly higher serum DcR3 levels compared to that without lymph node metastasis and systemic metastasis(P<0.01,P<0.05).But the levels of serum DcR3 had no correlation with differentiation grades and infiltrate depth(P>0.05,P>0.05).Conclusions Serum DcR3 levels were correlated significantly with TNM stage.This can help to evaluate the pre-operative clinical classification and decide the optimal extent of lymph node and adjacent organs dissection for gastric carcinoma.

    Objective To investigate the expression of CD44v6 protein in breast cancer and its clinical significance. Method SP immunohistochemical method was used to assess the expression of CD44v6 protein in 77 mastopathy tissue and the results were correlated with the clinical and data hibtophathologk.Result The expressions of CD44v6 protein in 12 benign cases were all negative.The positive expression rate of CD44v6 protein was 64.6%in the total 65 breast cancer cases.The expression rate in the positive and negative axillary lymph nodes groups were 100%and 25.8%(P<0.05 ) rspectivey;In histopathologic gradeⅠ,ⅡandⅢ,the expression rates were 9.1%,61.8%and 100%,respectively.There were significant differences among the three groups(P<0.001).Logistic multivariate regression analysis showed that the factors affected the expression of CD44v6 were axillary lymph nodes metastasis,tumor size and histopathologic grade.Conclusion CD44v6 protein can be applied as an important indicator in evaluating the malignant transformation,metastases and prognosis of breast cancer.

    Objective To investigate the relationship between efficacy of chemotherapy and serum Her-2/neu concentration in patients with advanced breast cancer,and to analyze its clinical significance.Methods Serum Her-2/neu concentration before and after chemotherapy collected from 66 cases of advanced breast cancer were detected by ELISA and the results were compared with Her-2/neu expression in tissues.The correlation,the efficacy of chemotherapy and time to progress(TTP) were analyzed. Results Serum Her-2/neu concentration was consistent with the expression of Her-2/neu in tumor tissues of patient with advanced breast cancer(P<0.001).The level of Her-2/neu before chemotherapy had no relation to efficacy of chemotherapy (P>0.05).Mean serum Her-2/neu concentration decreased from 33.97ng/ml to 22.11ng/ml after treatment in patient who was effective to chemotherapy,indicating the correlation to response rate of chemotherapy(P<0.001).Conclusion The changes of serum Her-2/neu concentration are significantly associated with efficacy of chemotherapy.Her-2/neu may be used as an indicator in the course of treatment Elevated serum Her-2/neu concentration before chemotherapy was correlated to shorter overall survival.

Results Serum Her-2/neu concentration was consistent with the expression of Her-2/neu in tumor tissues of patient with advanced breast cancer(P<0.001).The level of Her-2/neu before chemotherapy had no relation to efficacy of chemotherapy (P>0.05).Mean serum Her-2/neu concentration decreased from 33.97ng/ml to 22.11ng/ml after treatment in patient who was effective to chemotherapy,indicating the correlation to response rate of chemotherapy(P<0.001).Conclusion The changes of serum Her-2/neu concentration are significantly associated with efficacy of chemotherapy.Her-2/neu may be used as an indicator in the course of treatment Elevated serum Her-2/neu concentration before chemotherapy was correlated to shorter overall survival.

    Objective To evaluate the practical value of ultrasound-guided mammotome minimally invasive peeling on benign breast disease.Methods 856 benign breast masses from 435 patients were treated by ultrasound-guided mammotome minimally invasive peeling.Results All of breast masses were resected safely and completely.Each mass was resected by 18 peelings on average,and the average operation time was 15 minutes.The maximum diameter of the masses measured 1.5±1.0cm,and the lengths of incision were only 3 to 5 mm.No serious complications occurred among the patients.Conclusion Ultrasound-guided mammotome minimally invasive peeling for benign breast disease appears to be a procedure for widespread using because of its simplicity,accuracy,safety and minimally invasiveness.

Accumulated survival rate and average survival time were estimated using Kaplan-Meier estimates and compared with the logrank test where indicated.Results Accumulated survival rate of patients whose 3 cycle postoperative chemotherapy serum CA_(125) were≤35U/ml had no significant difference(P = 0.0693) from that of CA_(125)>35U/ml.There was no significant difference(P = 0.1074) between the group of CA_(125)≤70U/ml and>70U/ml as well.Accumulated survival rate of patients whose 3 cycle postoperative chemotherapy serum CA_(125)≤105 U/ml was significantly higher than those whose CA_(125)>105U/ml(P = 0.0171). Conclusions 105U/ml of CA_(125) after 3 cycle postoperative chemotherapy could be applied to evaluate prognosis of epithelial ovarian carcinoma.

Paclitaxel or Docetaxel was administered intravenously at a dose of 135mg/m~2(or 75 mg/m~2)on d1,Cisplatin 30mg/m~2 from d1 to d3.A treatment cycle was 21 days.After 2 cycles,the pathologic parameters,the treatment efficacy and the prognosis were compared between the patients of TNBC and non-TNBC.Results 20 of 147 patients(13.6%) were TNBC.Patients of TNBC had higher overall response rate compared with non-TNBC(P = 0.048 ).The median progression free survial time in the patients of TNBC and non-TNBC were 5.5 months and 6.5 months respectively.There were significant difference between the two groups at primary tumor size(P =0.042).Conclusion Paclitaxel or Docetaxel combined with Cisplatin chemotherapy regimen was sensitive in TNBC,but the median progression free survial time was worse in this group.
